Open flow cytometer with the combination of 3D hydrodynamic single cell focusing and confocal laser-induced fluorescence detection

Chengxin Wu et al.

Summary: We constructed an open and low-cost flow cytometer that integrates single cell aligning and fluorescence detection functions. The hardware cost for the LIF detection unit and 3D focusing device is $3200 and $400 respectively. The flow cytometer showed favorable assay precision and accuracy, and was successfully applied for evaluating ROS generation in single HepG2 cells.

Cellular heterogeneity in disease severity and clinical outcome: Granular understanding of immune response is key

Kriti Khare et al.

Summary: Understanding cellular heterogeneity is crucial for determining the disease severity and clinical outcomes of COVID-19. This article reviews current findings on immune response dysregulation and impairment in different disease severity sub-phenotypes. It highlights the potential role of cellular heterogeneity in COVID-19 severity and emphasizes the importance of factors such as age, gender, comorbidities, and genetics in regulating disease severity. The article also discusses the challenges and potential of single-cell genomics in studying heterogeneity for future pandemic preparedness.

With great power comes great responsibility: high-dimensional spectral flow cytometry to support clinical trials

Megan McCausland et al.

Summary: Flow cytometry is a powerful technology used for cell identification and characterization in research, drug development, and clinical sample analysis. Recent advancements in instrumentation and fluorochrome availability have led to increases in complexity and dimensionality of flow cytometry panels, presenting challenges in panel design, optimization, and downstream data analysis and interpretation. In this paper, practices for developing high-dimensional panels on the Aurora spectral flow cytometer to aid clinical sample analysis are described.

Full Spectrum Flow Cytometry as a Powerful Technology for Cancer Immunotherapy Research

Diana L. Bonilla et al.

Summary: The focus is on understanding the reasons for checkpoint blockade failure and identifying immunological biomarkers that can predict treatment response, disease progression, or adverse effects. Flow cytometry, as a single-cell analytical tool, is used to monitor various types of cancer, but the conventional flow cytometry no longer meets the need to evaluate multiple features simultaneously, thus prompting the adoption of Full Spectrum Flow Cytometry (FSFC).
