Optimizing efficient PCR-amplifiable DNA extraction from herbarium specimens

Fred E. Gouker et al.

Summary: The objective of this study was to optimize a DNA extraction protocol for obtaining high-quality DNA from preserved herbarium tissue. The improved protocol yielded DNA comparable in quality to commercially available kits and suitable for other applications. This simple and fast method utilizes standard laboratory-grade chemicals.

Optimizing the lysis step in CTAB DNA extractions of silica-dried and herbarium leaf tissues

S. James Carey et al.

Summary: The use of cetyltrimethylammonium bromide (CTAB) is an effective and inexpensive method for extracting DNA from plants. Modifying the CTAB protocol can optimize DNA extractions, where chemical additives, incubation temperature, and lysis duration have been found to affect DNA quantity and quality. Silica gel-preserved tissues yield higher DNA quantity, longer DNA fragments, and purer extractants compared to herbarium-preserved tissues.

What is the modified CTAB protocol? Characterizing modifications to the CTAB DNA extraction protocol

John J. Schenk et al.

Summary: Cetyltrimethylammonium bromide (CTAB)-based methods are widely used for DNA isolation from plant tissues, but optimizations are necessary due to the unique chemical composition of secondary metabolites among plant species. Many research articles cite modified CTAB protocols without detailing the specific alterations, leading to non-reproducible studies. This review surveyed the literature for modified CTAB protocols and summarized the modifications to provide extraction optimization recommendations. Standardizing DNA extractions through the review of modifications and the provided protocols could improve the repeatability and transparency of future genomic studies.

Modified CTAB protocols for high-molecular-weight DNA extractions from ferns

Pei-Jun Xie et al.

Summary: This study presents two modified CTAB protocols for efficient extraction of high-molecular-weight (HMW) DNA from ferns and evaluates their effectiveness in various fern taxa. These methods minimize mechanical disruption during lysis to prevent DNA shearing, resulting in high-quality HMW DNA with high yield.

How do surfactants unfold and refold proteins?

Daniel E. Otzen et al.

Summary: Although the mode of interaction between the anionic surfactant sodium dodecyl sulfate (SDS) and proteins has been a subject of controversy, recent studies have conclusively supported the core-shell model, where denatured proteins cover the surface of SDS micelles. The formation of these structures depends on the SDS:protein ratio and molecular mass of the protein. SDS denaturation can be reversed by adding nonionic surfactants, and is generally reversible except under low pH conditions.

Ubiquitin forms conventional decorated micelle structures with sodium dodecyl sulfate at saturation

Henriette Gavlshoj Mortensen et al.

Summary: Research has shown that the anionic surfactant sodium dodecyl sulfate interacts strongly with most globular proteins, denaturing and unfolding them to form protein-decorated SDS micelles. Investigation of the SDS-UBI system through various techniques revealed that SDS gradually unfolds UBI and forms protein-decorated SDS micelles.
