A census of the lung: CellCards from LungMAP

Xin Sun et al.

Summary: The human lung plays important roles in respiration, host defense, and basic physiology. Recent technological advancements have led to the discovery of new cell types and functional properties in the lung. A comprehensive cellular census of the lung has been synthesized, with detailed information on cell types, function, markers, developmental lineages, heterogeneity, regenerative potential, disease links, and key experimental tools. This publication aims to serve as a guide for lung research and promote efforts to establish, maintain, and restore respiratory health.

Inflammatory blockade prevents injury to the developing pulmonary gas exchange surface in preterm primates

Andrea Toth et al.

Summary: Perinatal inflammatory stress can have detrimental effects on lung development and lead to lifelong consequences. Using a prenatal rhesus macaque model, this study demonstrated that inflammation disrupts the developing primate lung, causing damage to the gas exchange surface and simplification of the alveoli. Blocking inflammatory cytokines can ameliorate this inflammatory lung injury and restore structural integrity.

Endothelial progenitor cells stimulate neonatal lung angiogenesis through FOXF1-mediated activation of BMP9/ACVRL1 signaling

Guolun Wang et al.

Summary: The molecular mechanisms through which pulmonary endothelial progenitor cells stimulate lung angiogenesis have been revealed to be the activation of the BMP9/ACVRL1/SMAD1 signaling pathway by FOXF1 and FLI1 nuclear proteins. This discovery contributes to a better understanding of the regulation of lung angiogenesis.

DIRECT-NET: An efficient method to discover cis-regulatory elements and construct regulatory networks from single-cell multiomics data

Lihua Zhang et al.

Summary: The emergence of single-cell multiomics data provides unprecedented opportunities to study the transcriptional regulatory mechanisms controlling cell identity. DIRECT-NET, a machine-learning method based on gradient boosting, allows us to identify genome-wide cis-regulatory elements (CREs) and their relationship to target genes from single-cell data. DIRECT-NET substantially improves the accuracy of inferring CRE-to-gene relationships and reveals cell subpopulation-specific and dynamic regulatory linkages.

Fgf10/Fgfr2b Signaling Orchestrates the Symphony of Molecular, Cellular, and Physical Processes Required for Harmonious Airway Branching Morphogenesis

Matthew R. Jones et al.

Summary: The development of airway branching relies on the coordination of various biological and physical factors across different spatial scales, with the Fgf10/Fgfr2b signaling pathway playing a key role. While fine reviews on molecular mechanisms related to branching morphogenesis have been published, a comprehensive review covering all major factors involved in branching and the role of Fgf10/Fgfr2b is lacking. This review aims to summarize existing literature on airway branching morphogenesis, emphasizing the impact of biophysical and mechanical forces, extracellular matrix remodeling, and other pulmonary networks on the process.

Genomic, epigenomic, and biophysical cues controlling the emergence of the lung alveolus

Jarod A. Zepp et al.

Summary: Through a multimodal approach, AT1 epithelial cells were identified as a signaling hub in the transition to air breathing at birth, aligning with force-exerting myofibroblasts to drive alveolar formation. Biophysical forces and AT1-derived paracrine signals together drive the alignment of cell fates, leading to extensive tissue remodeling required for postnatal respiration.
