Targeting endothelial tight junctions to predict and protect thoracic aortic aneurysm and dissection

Summary: Single-cell RNA sequencing revealed abnormal expressions of endothelial tight junctions (TJs) in thoracic aortas of patients with TAAD. In a BAPN-induced TAAD mouse model, disruption of endothelial TJ function and decreased distribution of crucial TJ components were observed. MRI imaging using two dextrans conjugated with Gd-DOTA as contrast agents showed accumulation in the thoracic aortas of BAPN-fed mice, indicating the potential of using these probes as non-invasive indicators of endothelial TJ function and TAAD.

Endothelial dysfunction in Marfan syndrome mice is restored by resveratrol

Summary: This study investigates the properties of endothelial cells (ECs) in mice with Marfan syndrome. The researchers found that ECs in the aorta of Marfan syndrome mice had reduced alignment with blood flow, increased roundness, larger surface area, and decreased junctional linearity. These alterations were most prominent in the ascending aorta and occurred before aortic dilatation. Treatment with resveratrol restored normal EC morphology. This study highlights the involvement of ECs in the development of aortic pathology in Marfan syndrome and suggests ECs as potential targets for future pharmacotherapies.

The Genetics and Typical Traits of Thoracic Aortic Aneurysm and Dissection

Summary: Genetic predisposition and risk factors such as hypertension and smoking can lead to the development of thoracic aortic aneurysm (TAA). Recent studies have identified monogenic defects in multiple genes and genetic modifiers and risk-conferring polymorphisms associated with TAA. Advances in omics technology have provided insights into various processes related to aortic wall degeneration, including inflammation, epigenetics, smooth muscle phenotype change, reactive oxygen species generation, mitochondrial dysfunction, and angiotensin signaling dysregulation. These findings open up possibilities for developing therapies that can halt or even reverse aneurysm progression.
