RyR2 inhibition with dantrolene is antiarrhythmic, prevents further pathological remodeling, and improves cardiac function in chronic ischemic heart disease

Jeffrey Schmeckpeper et al.

Summary: This study investigates the importance of targeting RyR2 hyperactivity in chronic ischemic heart disease (CIHD) in order to suppress ventricular tachycardia (VT) inducibility and prevent further progression of heart failure. The RyR2 inhibitor dantrolene was found to reduce VT inducibility and prevent contractile dysfunction in CIHD mice.

Electrophysiology and Arrhythmogenesis in the Human Right Ventricular Outflow Tract

Kedar Aras et al.

Summary: The electrophysiology of human right ventricular outflow tract (RVOT) is characterized by shorter action potential duration and the arrhythmogenic effects of adrenergic stimulation can be attenuated by cholinergic stimulation. Additionally, arrhythmia wave fronts and rotors in RVOT are relatively more organized in the endocardium compared to the epicardium.

Simultaneous triple-parametric optical mapping of transmembrane potential, intracellular calcium and NADH for cardiac physiology assessment

Sharon A. George et al.

Summary: In this study, triple-parametric optical mapping was used to simultaneously assess multiple parameters in cardiac physiology and pathophysiology. The technique was applied to study the effects of drugs on cardiac function and to investigate metabolic and electrical changes during ischemia and reperfusion.

RYR2 Channel Inhibition Is the Principal Mechanism of Flecainide Action in CPVT

Dmytro O. Kryshtal et al.

Summary: The main mechanism of antiarrhythmic action of flecainide is likely through inhibition of RyR2-mediated arrhythmogenic Ca release, rather than solely through sodium channel block. Experimental results in cardiomyocytes and mouse models have both confirmed this.

Ryanopathies and RyR2 dysfunctions: can we further decipher them using in vitro human disease models?

Yvonne Sleiman et al.

Summary: Regulation of intracellular calcium homeostasis is crucial for normal cell functions, with abnormal RyR2 function implicated in various diseases. Animal and human cell models have contributed valuable insights into RyR2 dysfunctions, offering potential for personalized therapeutics.

Ventricular Arrhythmias in Ischemic Cardiomyopathy-New Avenues for Mechanism-Guided Treatment

Matthew Amoni et al.

Summary: Ischemic heart disease is a common cause of lethal ventricular arrhythmias and sudden cardiac death, with implantable cardioverter defibrillators and ablation therapy being the most effective treatments. Advances in mechanistic insights and technological innovation have led to improved interventional approaches, while pharmacological management still needs improvement.
