Human Xylosyltransferase I-An Important Linker between Acute Senescence and Fibrogenesis

Molecular Mechanisms of Cellular Senescence in Neurodegenerative Diseases

Summary: Neurodegenerative diseases, such as Alzheimer's and Parkinson's, are characterized by neuronal loss, protein aggregation, and chronic inflammation. Aging is a critical risk factor for these diseases, but the mechanism is not well understood. Cellular senescence, which involves changes in cellular phenotypes and increased secretion of certain molecules, is discussed in this review as a contributing factor to brain aging and neurodegenerative diseases. The potential of senescence modifiers and senolytics in treating these diseases is also explored.

Heterogeneity of Cellular Senescence: Cell Type-Specific and Senescence Stimulus-Dependent Epigenetic Alterations

Summary: The aim of this study was to characterize DNA methylation patterns in replicative and induced senescence in different human cell types. Three epigenetic signatures were identified: cell type- and treatment-specific signature, cell type-specific senescence-related signature, and cell type-transversal replicative senescence-related signature. Cluster analysis revealed distinct DNA methylation patterns in replicative senescent cells, and enrichment in pathways related to the nervous system was shown. Despite no statistically significant evidence of age acceleration, a trend of increased biological age in replicative senescent cultures of all three cell types was observed. This work highlights the heterogeneity of senescent cells and their impact on tissue homeostasis.

Absence of HDAC3 by Matrix Stiffness Promotes Chromatin Remodeling and Fibroblast Activation in Idiopathic Pulmonary Fibrosis

Summary: Idiopathic pulmonary fibrosis (IPF) is a chronic and fatal disease characterized by progressive and irreversible lung scarring associated with fibroblast activation. This study investigates the role of HDAC3, a histone deacetylase, in chromatin remodeling and gene expression in IPF-derived fibroblasts. The results show that HDAC3 is decreased in the nucleus of IPF fibroblasts and its inhibition leads to increased expression of profibrotic genes. The study also suggests that matrix stiffness, mimicked by hydrogels, promotes the loss of nuclear HDAC3 and worsens the fibrotic microenvironment. These findings indicate that HDAC3 could be a potential target for therapeutic interventions in IPF.

Targeting extracellular matrix glycation to attenuate fibroblast activation

Summary: In this study, the researchers found that accumulated AGEs in the ECM can alter the phenotype of fibroblast within a three-dimensional collagen matrix, leading to the acquisition of a cancer-associated fibroblast (CAF)-like phenotype. These effects can be blocked by neutralizing antibodies against the AGE receptor (RAGE) or by inhibiting focal adhesion signaling. In addition, an AGE cross-link breaker called ALT-711 can reduce the transformation of fibroblasts into the CAF-like phenotype. Decreased matrix stiffness also attenuates fibroblast activation. These findings suggest potential therapeutic strategies targeting non-enzymatic ECM glycation to regulate fibroblast activation.

Summary: In this study, the effect of matrix stiffness on nucleus pulposus (NP) cell senescence was investigated using polyvinyl alcohol (PVA) hydrogel with different stiffness to mimic the matrix stiffness of normal and degenerated NP tissue. The results showed that increased matrix stiffness induced NP cell senescence and increased cellular Young's modulus, possibly through upregulation of integrin beta 1 and p38 MAPK. Restoration of extracellular matrix synthesis inhibited the matrix stiffness-induced senescence of NP cells. These findings provide new insights into the pathological mechanism of intervertebral disc degeneration.

Summary: The study identified integrin alpha 3 as a key regulator in mediating interactions between degenerative NP cells and laminin-111, with blocking this integrin resulting in decreased cell attachment, reduced expression of phenotypic markers, and alterations in cytoskeletal organization. Additionally, integrin alpha 3 plays a critical role in how NP cells sense and alter phenotype in response to culture upon laminin.
