Multisite Assessment of Optical Genome Mapping for Analysis of Structural Variants in Constitutional Postnatal Cases

M. Anwar Iqbal et al.

Summary: This study compared optical genome mapping (OGM) with current standard-of-care (SOC) methods in clinical cytogenetics. OGM showed high concordance with SOC for technical and clinical classification, as well as reproducibility. The authors recommend OGM as an alternative to SOC for rapid detection and diagnosis in postnatal constitutional disorders.

Molecular characterization and investigation of the role of genetic variation in phenotypic variability and response to treatment in a large pediatric Marfan syndrome cohort

Josephina A. N. Meester et al.

Summary: In this study, a large cohort of pediatric patients with Marfan syndrome (MFS) was examined to investigate the association between FBN1 variants and clinical characteristics and treatment response. It was found that (likely) pathogenic FBN1 variants were present in 91% of the patients. The study identified important genotype-phenotype associations and confirmed existing ones, which have implications for prognostic counseling of families with MFS.

Optical genome mapping enables constitutional chromosomal aberration detection

Tuomo Mantere et al.

Summary: The study investigated the use of optical genome mapping (OGM) to detect known chromosomal aberrations, with results showing OGM's ability to accurately detect various types of chromosomal abnormalities, including complex rearrangements. High-resolution OGM achieved 100% concordance compared to standard assays for aberrations without centromeric breakpoints.
