Specialized Pro-Resolving Mediators as Resolution Pharmacology for the Control of Pain and Itch

Summary: Specialized pro-resolving mediators (SPMs) are endogenous lipid mediators synthesized from omega-3 polyunsaturated fatty acids during inflammation. Synthetic SPMs have shown potent inflammation-resolving effects and analgesia in preclinical models, including rodent models of different types of pain. SPMs act through multiple G protein-coupled receptors expressed by immune cells, glial cells, and neurons, and their loss can impair pain resolution. SPMs also have an emerging role in itch control, and targeting SPMs or SPM receptors pharmacologically could lead to novel therapeutics for pain and itch.

The results showed that inhibiting CYP2J2 can enhance the inhibitory effect of ω-3 LCPUFA on CNV, and co-treatment with montelukast, which inhibits CYP2C8, further enhances the suppression of CNV.
