6-Shogaol inhibits the proliferation, apoptosis, and migration of rheumatoid arthritis fibroblast-like synoviocytes via the PI3K/AKT/NF-κB pathway

Nan Li et al.

Summary: This study found that 6-shogaol inhibits proliferation, migration, and invasion, and induces apoptosis in rheumatoid arthritis fibroblast-like synoviocytes (FLSs). It also reduces the production of inflammatory cytokines and matrix metalloproteinases (MMPs). Molecular mechanism analysis revealed that 6-shogaol inhibits the PI3K/AKT/NF-kappa B pathway by activating PPAR-gamma. Treatment with 6-shogaol ameliorated joint destruction in a mouse model of collagen-induced arthritis (CIA). Therefore, 6-shogaol may be a potential natural drug for future rheumatoid arthritis treatments.

Astragaloside IV as a novel CXCR4 antagonist alleviates osteoarthritis in the knee of monosodium iodoacetate-induced rats

Kuangyang Yang et al.

Summary: In this study, the researchers found that Astragaloside IV (ASN IV), a phytochemical found in Astragalus membranaceus, acts as a novel CXCR4 antagonist, protecting cartilage in osteoarthritis (OA) patients. ASN IV inhibits the overexpression of ADAMTS4,5 in chondrocytes by blocking Akt signaling pathway. Furthermore, ASN IV administration repairs damaged cartilage and subchondral bone in an animal model of OA. This study suggests that repurposing ASN IV as a joint protective agent could be a promising strategy for OA drug development.

Tanshinone IIA relieves arthritis by inhibiting autophagy of fibroblast-like synoviocytes via matrix metalloproteinase9/receptor for advanced glycation end product/toll-like receptor 9 signal axis in mice with collagen-induced arthritis

Wei Liu et al.

Summary: In this study, the researchers investigated the mechanisms by which Tanshinone IIA (TIIA) affects RA fibroblast-like synoviocytes (RA-FLSs). They used a mouse model of RA called collagen-induced arthritis (CIA) and manipulated matrix metalloproteinase9 (MMP9), receptor for advanced glycation end product (RAGE), and toll-like receptor 9 (TLR9) in both mice and RA-FLSs to study the effects of TIIA. The results showed that TIIA inhibited the proliferation of RA-FLSs, affected autophagy, and improved the symptoms of RA in mice by inhibiting the expression of MMP9 and the activation of TLR9 via the RAGE pathway.

Summary: Research has shown that the Chinese herbal formula Longteng Decoction (LTD) can inhibit joint inflammation, reduce joint pain and swelling, shorten morning stiffness, decrease ESR, and suppress joint inflammation in the treatment of rheumatoid arthritis (RA). Furthermore, LTD can modulate the ILC2s immune response network to slow down the inflammatory process of synovial tissue, making it a potential adjunct or alternative therapy for RA.

Inhibition of MAGL activates the Keap1/Nrf2 pathway to attenuate glucocorticoid-induced osteonecrosis of the femoral head

Ning Yang et al.

Summary: Glucocorticoid-induced osteonecrosis of the femoral head (ONFH) remains poorly understood, with studies suggesting that monoacylglycerol lipase (MAGL) in the endocannabinoid system plays a crucial role in regulating oxidative stress and BMSC fate. Inhibition of MAGL can reduce oxidative stress, rescue BMSC apoptosis, and effectively attenuate GC-induced ONFH through the Keap1/nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Therefore, MAGL may be a promising target for prevention and treatment of GC-induced ONFH.
