Due to their diverse medicinal properties, isoxazole derivatives have great potential in the development of novel drugs with enhanced effectiveness and lower harm.

Enantioselective Construction of Consecutive Tetrasubstituted Stereogenic Centers by Reaction of ?-Substituted ?-Nitroacrylates with Oxazol-5-(4H)-ones Catalyzed by Cinchona Alkaloid Sulfonamide Catalysts

Summary: The enantioselective reaction of a-substituted ig-nitroacrylates with oxazol-5-(4H)-ones (oxazolones) to construct consecutive tetrasubstituted stereogenic centers was achieved. A cinchona alkaloid sulfonamide catalyst provided products with vicinal chiral centers that exhibited excellent enantio- and diastereoselectivities. The obtained products were further transformed into different chiral compounds without compromising their enantiopurity. Moreover, density functional theory (DFT) calculations were employed to elucidate the mechanism and origin of the observed stereoselectivity of the reaction.

A New Organocatalytic Desymmetrization Reaction Enables the Enantioselective Total Synthesis of Madangamine E

This reaction efficiently constructed a chiral bicyclic core with near-perfect enantio- and diastereo-selectivity, establishing multiple stereogenic centers, including a quaternary carbon, and demonstrating high scalability. Following the construction of the bicyclic core, the total synthesis of madangamine E was completed with key steps including oxidative lactamization, olefination, and ring-closing metatheses.
