Extended Work Function Shift of Large-Area Biofunctionalized Surfaces Triggered by a Few Single-Molecule Affinity Binding Events

Summary: Kelvin probe force microscopy (KPFM) imaging shows that a small number of binding events on a large-area Au surface biofunctionalized with approximately 10^8 capturing antibodies can cause an extended work function change. An incubation of 10 minutes in a phosphate buffer saline solution containing approximately 10 IgM antigens produces a work function shift of approximately -60 meV. This shift involves the entire inspected area and is in agreement with the threshold voltage shift of an electrolyte-gated single-molecule large-area transistor (SiMoT).
