The Risk Factors and Clinical Features of Acanthamoeba Keratitis: First Time Detection of Acanthamoeba T5 Genotype from Keratitis Patients in Turkey

Summary: This study aimed to investigate the presence and genotypes of Acanthamoeba in clinical samples of keratitis patients and in samples from asymptomatic contact lens users. Acanthamoeba was detected in a small percentage of keratitis patients, with the main genotypes being T4 and T5. However, no Acanthamoeba was found in samples from asymptomatic contact lens users.

Genotyping and Molecular Identification of Acanthamoeba Genotype T4 and Naegleria fowleri from Cerebrospinal Fluid Samples of Patients in Turkey: Is it the Pathogens of Unknown Causes of Death?

Summary: This study investigated the presence of pathogenic free-living amoebae (FLA) in suspected cases of meningoencephalitis with unknown causes of death in Turkey using molecular diagnostic tools. The study identified the presence of Acanthamoeba genotype T4 and N. fowleri genotype II in cerebrospinal fluid (CSF) samples using real-time PCR assay. The findings highlight the importance of considering primary amoebic meningoencephalitis (PAM) and granulomatous amoebic encephalitis (GAE) as differential diagnoses in suspected meningoencephalitis or cases of unknown cause in Turkey. However, the use of real-time PCR has limitations.

Summary: A survey in 100 public freshwater sources across 28 provinces in Thailand identified 9 genotypes of Acanthamoeba, with T4 being the most predominant. The nucleotide diversity within local populations was significantly less than global isolates, and genotype T11 showed a higher level of nucleotide diversity possibly due to cryptic subtypes. The evolution of the 18S rRNA gene of the pathogenic-free living amoeba seems ongoing, suggesting there may be more genotypes yet to be discovered.
