A 14-day pulse of PLX5622 modifies & alpha;-synucleinopathy in preformed fibril-infused aged mice of both sexes

Summary: Reactive microglia are observed in aging and Lewy body disorders, and resetting these cells with the CSF1R inhibitor PLX5622 may have therapeutic potential. In the PFF model of alpha-synuclein fibrils, short-term dietary exposure to PLX5622 reduced inclusion numbers and insoluble alpha-synuclein in aged males, but raised inclusion sizes in both sexes. Transient delivery of PLX5622 also improved spatial reference memory in PFF-infused aged mice.

A postmortem study suggests a revision of the dual-hit hypothesis of Parkinson's disease

Summary: The dual-hit hypothesis of Parkinson's disease suggests that a neurotropic pathogen leads to formation of alpha-synuclein pathology that enters the brain from the olfactory bulb and the dorsal motor nucleus of the vagus. However, a re-analysis of postmortem datasets found that early body-first Lewy body disease (LBD) cases rarely had olfactory bulb pathology, while early brain-first LBD cases almost always showed olfactory bulb pathology. This supports the idea that the pathological process starts in either the olfactory bulb or the enteric nervous system, but not both simultaneously.

Summary: This study reveals a potential sex-biased link between alpha-synucleinopathy and APE1 in mice and humans, suggesting that APE1 may play a role in the formation of alpha-synuclein inclusions. Further studies are needed to understand how APE1 modifies alpha-synuclein inclusions and how alpha-synucleinopathy and biological sex interact to modify APE1.

Summary: Several staging systems exist for the neuropathological diagnosis of LBD, with varying levels of inter-rater reliability. The LPC system, based on a dichotomous approach for scoring LP, showed good reproducibility and allowed for the classification of all cases into distinct categories. It may serve as a reliable and useful standard future approach for the post-mortem evaluation of LP.

Heat Shock Protein 70 as a Sex-Skewed Regulator of α-Synucleinopathy

Summary: The study revealed gender differences in the role of Hsp70 in proteinopathic disorders, with exogenous Hsp70 reducing alpha-synuclein inclusions in cells of both sexes. Intranasal delivery of eHsp70 in male mice decreased disease progression, while no eHsp70 uptake into the brain was observed in females. Inducible Hsp70 expression was higher in amygdala tissues of male subjects, while women expressed higher levels of amygdalar Hsp70 regardless of disease status.
