A synergistic strategy of dual-crosslinking and loading intelligent nanogels for enhancing anti-coagulation, pro-endothelialization and anti-calcification properties in bioprosthetic heart valves

Dialdehyde xanthan gum and curcumin synergistically crosslinked bioprosthetic valve leaflets with anti-thrombotic, anti-inflammatory and anti-calcification properties

Summary: Currently, commercial glutaraldehyde (GA)-crosslinked bioprosthetic valve leaflets (BVLs) face problems such as thromboembolic complications, calcification, and limited durability. This study introduces a collaborative strategy using double crosslinking agents to enhance the mechanical and structural stability, hemocompatibility, and anti-calcification properties of BVLs. The strategy involves crosslinking decellularized porcine pericardium (D-PP) with dialdehyde xanthan gum (AXG) and curcumin to obtain AXG + Cur-PP, which showed improved anti-thrombotic properties and endothelialization potential, milder immune responses, and excellent anti-calcification properties compared to GA-crosslinked PP.

A bioprosthetic heart valve prepared by copolymerization of 2-isocyana- toethyl methacrylate modified pericardium and functional monomer

Summary: This study developed a new glutaraldehyde-free crosslinking method to prepare a poly(ethylene glycol) dimethacrylate (PEGDA) polymer crosslinked porcine pericardium (PICM-PEGDA-PP) with improved cytocompatibility, stability, mechanical properties, and anticalcification property compared to glutaraldehyde crosslinked porcine pericardium (GA-PP). The PICM-PEGDA-PP demonstrated low protein, platelet, and blood cell adsorption as well as significant antithrombotic performance. It meets the requirements for BHVs according to ISO 5840-3 and has the potential to be a clinical alternative for glutaraldehyde crosslinked BHVs in the future.

Sodium lignosulfonate cross-linked bioprosthetic heart valve materials for enhanced cytocompatibility, improved hemocompatibility, and reduced calcification

Summary: This study aimed to enhance the performance of bioprosthetic heart valves (BHV) by cross-linking porcine pericardium (PP) with sodium lignosulfonate (SLS). The results showed that SLS-crosslinked PP (SLS-PP) had improved cell proliferation, reduced protein adhesion, and decreased calcification compared to glutaraldehyde-crosslinked PP (Glut-PP). SLS is suggested as a potential alternative cross-linker for BHVs.

The effect of decellularisation on the real time mechanical fatigue of porcine aortic heart valve roots

Summary: This preliminary study investigated the influence of low concentration SDS decellularisation treatment on the mechanical fatigue of heart valve roots. A real time in vitro method was developed to compare decellularised and cellular valve roots, and assess the effects of fatigue on hydrodynamic function and mechanical properties. The results demonstrated that decellularised heart valve roots can withstand physiological cyclic deformations while maintaining their overall functionality and mechanical properties.

This study focused on the controlled delivery of antibiotic drugs using chitosan crosslinked dialdehyde xanthan gum interpenetrated hydroxypropyl methylcellulose gels, demonstrating promising results in terms of drug release kinetics, cell cytocompatibility, and antibacterial activity against Gram-negative and positive bacteria. Overall, the produced hydrogel scaffolds showed potential for controlled antibiotic delivery and microbial growth inhibition.
