Nanoparticles with ultrasound-induced afterglow luminescence for tumour-specific theranostics

A Hierarchical Structured Fiber Device Remodeling the Acidic Tumor Microenvironment for Enhanced Cancer Immunotherapy

Summary: A hierarchically structured fiber device is developed for local drug delivery to remodel the acidic tumor microenvironment and enhance the effectiveness of immunotherapy. Proton pump inhibitors in the fiber matrix inhibit the efflux of intracellular H+ from tumor cells, leading to remodeling of the acidic TME. Targeted micelles and M1 macrophage membrane-coated nanoparticles induce immunogenic cell death of tumor cells and phenotypic transformation of tumor-associated macrophages, respectively. The restoration of TME acidity promotes immunogenic cell death and polarization of macrophages, alleviating the immunosuppressive microenvironment and enhancing the antitumor immune response.

This nanosystem efficiently depletes YTHDF1 expression, suppresses cancer progression and metastasis, and enhances immune responses. The CD47 expression on the sEVs enhances phagocytosis of tumor cells by tumor-associated macrophages.

Rational Design of Platinum-Bismuth Sulfide Schottky Heterostructure for Sonocatalysis-Mediated Hydrogen Therapy

Summary: Conventional sonodynamic therapy is limited by the tumor microenvironment, so a concept of sonocatalytic hydrogen evolution is proposed as an oxygen-independent therapeutic approach. Using the narrow-bandgap semiconductor bismuth sulfide (Bi2S3) as the sonocatalyst, in situ grown platinum (Pt) nanoparticles are able to optimize their catalytic performance. The proposed therapy combines the generation of H2 and the depletion of glutathione (GSH) to induce tumor cell apoptosis while also relieving tumor hypoxia and reversing the immunosuppressive microenvironment.

ZIF-8 Nanoparticles Evoke Pyroptosis for High-Efficiency Cancer Immunotherapy

Summary: Researchers discovered that ZIF-8 nanoparticles can induce pyroptosis, a form of cell death, through a caspase-1/gasdermin D (GSDMD)-dependent pathway. This cell death process is accompanied by necrosis and immunogenic cell death (ICD), which triggers an immune response. Furthermore, loading a mitochondrial depolarizing agent, CCCP, into ZIF-8 nanoparticles enhances the pyroptosis process. The modified F127-ZIF-8(CCCP) nanoparticles activate anti-tumor immune response and reshape the tumor microenvironment, leading to effective tumor growth inhibition. This study advances the biomedical applications of ZIF-8 and offers insights into pyroptosis-induced cancer therapy.

A Polymeric Extracellular Matrix Nanoremodeler for Activatable Cancer Photo-Immunotherapy

Summary: Cancer immunotherapy has potential in the clinic, but the extracellular matrix (ECM) in the tumor microenvironment presents a barrier. A semiconducting polymer-based ECM nanoremodeler (SPNcb) is reported to combine photodynamic antitumor activity with the inhibition of collagen-crosslinking enzymes for cancer photo-immunotherapy. SPNcb blocks collagen crosslinking and decreases ECM stiffness, improving the tumor infiltration of cytotoxic T cells and inhibiting tumor growth and metastasis.
