Single-cell RNA sequencing reveals immunosuppressive myeloid cell diversity during malignant progression in a murine model of glioma

Summary: Recent studies have shown that the dynamic tumor microenvironment (TME) is important in high-grade gliomas (HGGs). It is still unclear whether myeloid cells play a role in the malignant progression of low-grade glioma (LGG). In this study, using single-cell RNA sequencing, the researchers investigated the cellular heterogeneity of the TME in a murine glioma model that mimics the progression from LGG to HGG. They found that LGGs have increased infiltrating T cells and natural killer cells in the TME, while HGGs lack this infiltration. The study also identified distinct macrophage clusters in the TME that exhibit an immune-activated phenotype in LGG but then transition to an immunosuppressive state in HGG.

Blood-brain barrier-penetrating single CRISPR-Cas9 nanocapsules for effective and safe glioblastoma gene therapy

Our nanocapsules can penetrate the blood-brain barrier, target tumor cells, and selectively release the Cas9/sgRNA complex. Treatment with nanocapsules significantly extended survival time.

Encapsulating Cas9 into extracellular vesicles by protein myristoylation

Summary: CRISPR/Cas9 genome editing is a promising method for treating genetic diseases. However, encapsulating the CRISPR/Cas9 machinery for delivery is challenging. In this study, the researchers found that myristoylated proteins were preferentially encapsulated into extracellular vesicles (EVs). By fusing a myristoylated peptide onto the N-terminus of Cas9, they were able to promote the myristoylation and encapsulation of Cas9 into EVs. Additionally, introducing VSV-G increased the transfection potential. The encapsulated Cas9 accounted for 0.7% of total EV protein. The EVs coated with VSV-G encapsulating Cas9/sgRNA-eGFP showed up to 42% eGFP knock out efficiency in recipient cells, with limited off-target effects.
