Engineering ROS-Responsive Bioscaffolds for Disrupting Myeloid Cell-Driven Immunosuppressive Niche to Enhance PD-L1 Blockade-Based Postablative Immunotherapy

Shaoyue Li et al.

Summary: Inadequate ablation can result in an immunosuppressive microenvironment dominated by myeloid cells, leading to resistance to immune checkpoint blocking therapy. This study developed a scaffold system that delivers IPI549 and aPDL1 to disrupt the immunosuppressive microenvironment and enhance anti-tumor immune response. Preclinical models demonstrated that this biomaterial system mimics a hot tumor-immunity niche, inhibiting tumor progression and metastasis, and protecting cured mice against tumor rechallenge.

Gold-seaurchin based immunomodulator enabling photothermal intervention and aCD16 transfection to boost NK cell adoptive immunotherapy

Xinyi Lin et al.

Summary: This study explores a nano-immunomodulator, AuNSP@alpha CD16, to improve the antitumor effects of adoptive NK cells by remodeling the tumor microenvironment. The AuNSP can trigger mild photothermal intervention to partially destroy tumors and collapse physical barriers, allowing infiltration of NK cells. It can also modify tumor surface with CD16 antibody, leading to strong NK cell activation. The designed AuNSP@alpha CD16 induces an immune-favorable tumor microenvironment, enhancing the killing functions of NK cells against solid tumors.

Improvement of the anticancer efficacy of PD-1/PD-L1 blockade via combination therapy and PD-L1 regulation

Mengling Wu et al.

Summary: PD-1/PD-L1 immune checkpoints inhibit the anticancer effect of T cells in the tumor microenvironment, and downregulation or upregulation of PD-L1 expression can improve the efficacy of anti-PD-1/PD-L1 treatment. Overcoming the current limitations of this treatment can expand its anticancer applications and improve patient response rates and survival time.

Down-regulation of MLLT1 super elongation complex subunit impairs the anti-tumor activity of natural killer cells in esophageal cancer

Chong Liu et al.

Summary: This study reveals that the expression of MLLT1 protein is down-regulated in NK cells of patients with esophageal cancer, leading to a decrease in their cytotoxicity. This down-regulation is associated with the TIM-3 signaling pathway, suggesting that MLLT1 could be a potential target for future NK cell-mediated immunotherapy against esophageal cancer.
