Behavioral, Anti-Inflammatory, and Neuroprotective Effects of a Novel FPR2 Agonist in Two Mouse Models of Autism

Summary: Autism spectrum disorders (ASD) are a group of neurodevelopmental conditions characterized by social deficits, repetitive behaviors, and altered inflammatory responses. This study found that ASD children have decreased levels of lipoxin A4 (LXA4), a mediator involved in inflammation resolution. Activation of the receptor FPR2 by a compound called MR-39 had positive effects on inflammation markers, LXA4 expression, and social behavior in validated animal models of ASD. These findings suggest that FPR2 could be a potential target for therapeutic approaches in ASD.

Exogenous Annexin 1 inhibits Th17 cell differentiation induced by anti-TNF treatment via activating FPR2 in DSS-induced colitis

Summary: This study discovered that inhibition of Th17 cells is significant for improving the effectiveness of anti-TNF treatment in Inflammatory Bowel Disease (IBD). By using the anti-TNF nanobody V7 and ANX1 to treat DSS-induced colitis mice, it was found that V7 induces Th17 cell differentiation through phagocytosis by DCs, while ANX1 inhibits Th17 cell differentiation by activating the FPR2-STAT3 pathway. Combination therapy with V7 and ANX1 showed better therapeutic effects in improving DSS-induced colitis.

It concludes that there is limited biosynthetic capacity for SPMs in human leukocytes, the identity and signaling of proposed SPM receptors need further validation, SPM levels are not related to dietary supplementation or resolution of inflammation, and the commonly reported methodology for quantification of SPMs is unreliable.

Stimulus-specific remodeling of the neuronal transcriptome through nuclear intron-retaining transcripts

Summary: The nuclear envelope not only acts as a physical barrier between the nucleus and cytosol, but also plays a role in regulating gene expression. This study reveals that a majority of stable intron-retaining mRNAs are retained in the nucleus in mature mouse cortical neurons. Upon neuronal stimulation, some transcripts undergo degradation while others complete splicing and are exported to the cytosol, leading to rapid increases in protein levels. Different forms of stimulation mobilize distinct groups of intron-retaining transcripts, with selectivity arising from specific signaling pathways.
