Computational investigation of aphid odorant receptor structure and binding function

Native Function of the Bacterial Ion Channel SthK in a Sparsely Tethered Lipid Bilayer Membrane Architecture

Tethered bilayer lipid membranes (tBLMs) provide a suitable model system to study membrane proteins in isolation. Putative ligand binding sites of two functionally characterized bark beetle odorant receptors

Summary: This study annotated 73 ORs from an antennal transcriptome of I. typographus and reported the functional characterization of two ORs (ItypOR46 and ItypOR49) responsive to single enantiomers of common bark beetle pheromone compounds. Homology modeling and molecular docking were used to predict their binding sites, providing insight into ligand-OR interaction for the functional evolution of the insect OR gene family. The ecological importance of these receptors and their potential use in pest control and biosensors to detect bark beetle infestations suggest further evaluation of these ORs.

Highly sensitive VOC detectors using insect olfactory receptors reconstituted into lipid bilayers

Multiscale modeling shows that dielectric differences make NaV channels faster than KV channels

Summary: The generation of action potentials in excitable cells relies on the activation kinetics of voltage-gated Na (Na-V) and K (K-V) channels, with Na-V channels activating much faster. Recent experimental evidence shows that the difference in activation kinetics is due to the presence of a highly conserved threonine in Na-V channels and isoleucine in K-V channels within the gating pore of the voltage sensor domain. Through multiscale modeling, it has been determined that the faster activation kinetics of Na-V channels is attributed to a stronger dielectric polarization effect by threonine compared to isoleucine.

Insect odorant receptor nanodiscs for sensitive and specific electrochemical detection of odorant compounds

Summary: This study presents an electrochemical sensing methodology using an insect odorant receptor (iOR) as the biological recognition element for detection of odorant compounds. The fruit fly Drosophila melanogaster's Or22a was used as a model receptor and shown to be functionally active towards its known ligand (ethyl hexanoate). The EIS sensor exhibited high sensitivity and specificity towards ethyl hexanoate with a detection limit of 5.5 fM, with neutron reflectometry studies providing evidence of conformational changes in the receptor upon ligand binding.

The structural basis of odorant recognition in insect olfactory receptors

The olfactory receptor MhOR5 from the jumping bristletail is a homotetrameric odorant-gated ion channel with broad chemical tuning, recognizing odorants through distributed hydrophobic interactions in a simple binding pocket. The receptor's sensitivity to odorants can be predictably altered by mutating individual residues in the binding pocket, shedding light on the molecular recognition mechanisms of the olfactory system.
