This review highlights recent advances in cancer-derived exosomal miRNAs, including their sorting and delivery mechanisms, as well as their potential as diagnostic and prognostic markers and in detecting cancer resistance to therapeutic agents.

Exosomal microRNAs array sensor with a bioconjugate composed of p53 protein and hydrazine for the specific lung cancer detection

Summary: This study demonstrates a novel strategy for the early diagnosis of lung cancer by detecting microRNAs encapsulated in exosomes using immunomagnetic isolation. The researchers selectively extracted exo-miRNAs from patient serum by capturing miRNA from lysed exosomes with specially designed capture probe modified magnetic beads. The sensor probe formed through T4 DNA polymerase-mediated in situ formation of chimeric 5'-miRNA-DNA-3' (Target) and p53 protein recognition showed excellent analytical performance, with a dynamic range between 100 aM and 10 pM and a detection limit of 92 (+/- 0.1) aM. Furthermore, a multiplexed array sensor was prepared to detect four exo-miRNAs at femtomolar levels in cell culture media and lung cancer patient serum samples.

A tri-channel electrochemical immunobiosensor has been developed for enzyme-free and label-free detection of carcino-embryonic antigen (CEA), neuron-specific enolase (NSE), and cytokeratin 19 fragments (Cyfra21-1) from exosomes for specific early diagnosis of lung cancer. The electrochemical immunobiosensor showed good selectivity and stability, with a detection limit down to 10(-4) ng/mL. Different expression levels of exosomal markers were observed in different types of lung cancer.
