Exploring the HSA/DNA/lung cancer cells binding behavior of p-Synephrine, a naturally occurring phenyl ethanol amine with anti-adipogenic activity: multi spectroscopic, molecular dynamic and cellular approaches

Reza Taheri et al.

Summary: This study investigates the interaction between p-Synephrine (SN), human serum albumin (HSA), and calf thymus DNA (ctDNA) using various biophysical methods and molecular dynamics simulation techniques. The results show that SN binds to HSA and ctDNA, leading to conformational changes, stability enhancement, and inhibition of lung cancer cell viability through the downregulation of the PI3K/AKT/mTOR signaling pathway.

Analysis of the Equilibrium Distribution of Ligands in Heterogeneous Media-Approaches and Pitfalls

Maria Joao Moreno et al.

Summary: This review article discusses various methods and formalisms for analyzing the equilibrium distribution of ligands in the presence of proteins, lipid membranes, or both, with special attention to common pitfalls and validity limits of different approaches. The analysis of ligand binding through Stern-Volmer plots of protein fluorescence quenching is emphasized for its widespread use in characterizing interactions. Different systems of increasing complexity are considered, from proteins with single to multiple binding sites to membrane proteins in lipid bilayers.

Biophysical Characterization of the Interaction between a Transport Human Plasma Protein and the 5,10,15,20-Tetra(pyridine-4-yl)porphyrin

Otavio Augusto Chaves et al.

Summary: In this study, the interaction between human serum albumin (HSA) and a non-charged synthetic photosensitizer was evaluated using spectroscopic techniques and molecular docking. The results showed a ground-state association between HSA and the photosensitizer, with no dynamic fluorescence quenching. The binding affinity was moderate and spontaneous, causing only weak perturbation on the secondary structure of albumin.

Fluorescent pyrene moiety in fluorinated C6F5-corroles increases the interaction with HSA and CT-DNA

Thiago Acunha et al.

Summary: Fluorinated corroles were biologically evaluated for their binding affinity to human serum albumin (HSA) and calf-thymus DNA (CT-DNA) using multiple spectroscopic techniques and molecular docking calculations. The pyrene moiety increased interactions with both HSA and CT-DNA, likely due to pyrene planarity, while the fluorinated corroles showed different interaction modes with HSA and CT-DNA.

PLIP 2021: expanding the scope of the protein-ligand interaction profiler to DNA and RNA

Melissa F. Adasme et al.

Summary: With the growth of protein structure data, the analysis of molecular interactions between ligands and their target molecules becomes increasingly important. PLIP, a tool for protein-ligand interaction profiling, has been successful in detecting and visualizing these interactions, with applications ranging from characterizing docking experiments to evaluating novel ligand-protein complexes. Additionally, PLIP has been extended to encompass interactions with DNA and RNA, demonstrating its utility in scenarios such as cancer drug targeting DNA and RNA-protein complexes associated with neurological diseases.
