Optic cup morphogenesis across species and related inborn human eye defects

Marcos J. Cardozo et al.

Summary: The vertebrate eye acquires its cup-like shape early in development through a process called optic cup morphogenesis, which has been studied using live imaging of transparent teleost embryos and mammalian stem cell-derived organoids. Molecular and pharmacological interventions have revealed the underlying molecular mechanisms and physical forces involved. This review summarizes the resulting scenarios and proposed models, including both common and species-specific events, and discusses how these studies can provide insights into human eye malformations resulting from failures in optic cup morphogenesis.

Recent advances in understanding the impact of infection and inflammation on hematopoietic stem and progenitor cells

Michael D. Milsom et al.

Summary: In this review article, the authors report on the progress made in understanding the interplay between infection, inflammation, and hematopoietic stem cells (HSCs) during the funding period of the Collaborative Research Center 873. The findings suggest that the proliferative response of HSCs to inflammatory cytokines may lead to HSC exhaustion and hematologic dysfunction.

Collective cell migration during optic cup formation features changing cell-matrix interactions linked to matrix topology

Karen G. Soans et al.

Summary: Cell migration is crucial for organismal development, but the influence of physical properties on migration phenomena in vivo remains unclear. In this study, the researchers used zebrafish optic cup formation to investigate the role of extracellular matrix (ECM) properties in cell migration. They found that rim cells migrate over an immobile ECM and require cryptic lamellipodia for movement. The topology of the ECM changes along the migration path and is accompanied by changes in cell-matrix interactions. Matrix porosity is linked to efficient migration.

Igf signaling couples retina growth with body growth by modulating progenitor cell division

Clara Becker et al.

Summary: In the neuroretina of medaka fish, the Igf1 receptor (Igf1r) plays a key role in proliferation control in the postembryonic retinal stem cell niche. Activation of Igf1r signaling can increase retinal size while preserving its structural integrity, indicating its importance as a key module for controlling retinal size and composition with evolutionary implications.

Phase transition specified by a binary code patterns the vertebrate eye cup

Revathi Balasubramanian et al.

Summary: The development of the vertebrate eye is determined by a combinatorial code of FGF and Wnt signaling, with FGF regulating the stem cell-like properties of the ciliary margin (CM) and Wnt maintaining a balance. FGF signaling promotes Wnt signaling in the CM through beta-catenin stabilization, while Wnt signaling converts the neural retina to either the CM or the retinal pigmented epithelium (RPE) depending on FGF signaling.

Meis homeobox genes control progenitor competence in the retina

Naoko Dupacova et al.

Summary: Meis1 and Meis2, through genetic ablation in the developing optic cup, maintain the retinal progenitor pool while suppressing the expression of CMZ and OD fates. Meis transcription factors bind regulatory regions of RPC, CMZ, and OD-specific genes, providing insight into the Meis-dependent gene regulatory network. Meis1 and Meis2 play an essential role as regulators of cell fate competence, organizing spatial territories in the vertebrate eye.

Build me up optic cup: Intrinsic and extrinsic mechanisms of vertebrate eye morphogenesis

Macaulie A. Casey et al.

Summary: The basic structure of the eye is established during embryonic development, with molecular genetics and imaging being used to quantify cellular processes such as adhesion and actomyosin activity. Studies reveal how intrinsic machinery and spatiotemporally specific extrinsic inputs collaborate to control dynamics of cell movements and morphologies. Recent advances in understanding eye morphogenesis focus on the mechanics of eye formation throughout vertebrate systems, including potential opportunities using organoids for testing hypotheses from embryonic models.

Retinal Stem Cell 'Retirement Plans': Growth, Regulation and Species Adaptations in the Retinal Ciliary Marginal Zone

Amanda Miles et al.

Summary: The vertebrate retina develops from specified precursor cells that generate lineages of neural retina, retinal pigmented epithelium, or ciliary body. In some species, proliferative stem-cell-like cells located at the peripheral margin, possibly representing a conserved retinal stem cell niche, continuously supply new retinal cells post-embryonically. This review highlights the research characterizing similar stem-like cells across different species and discusses their role in regulating retinal growth and plasticity.

Stretching of the retinal pigment epithelium contributes to zebrafish optic cup morphogenesis

Tania Moreno-Marmol et al.

Summary: The development of the vertebrate eye involves the reorganization of embryonic tissues to form a round shape, with the retinal pigment epithelium (RPE) playing a key role in this process. Studies in zebrafish have shown that RPE cells stretch and flatten to match the curvature of the retina, promoting the formation of the optic cup. This mechanism differs in species with slower development, such as mice and humans, where RPE expansion occurs through cell proliferation over a longer period of time.
