CMDB: the comprehensive population genome variation database of China

Zhichao Li et al.

Summary: The Chinese Millionome Database (CMDB) is a database that contains low-coverage whole-genome sequencing (WGS) data from 141,431 unrelated healthy Chinese individuals, covering 9.04 million single nucleotide variants (SNV) with allele frequency information. The CMDB is the most representative and comprehensive Chinese population genome database to date, housing data from a multi-ethnic Chinese population with wide geographical distribution.

Clinical Pharmacogenetics Implementation Consortium Guideline for CYP2C19 Genotype and Clopidogrel Therapy: 2022 Update

Craig R. Lee et al.

Summary: CYP2C19 enzyme plays a role in the activation of clopidogrel, and the genotype of CYP2C19 affects the formation of active metabolites of clopidogrel. Thus, using CYP2C19 genotype to guide the use of clopidogrel can reduce the risk of reduced platelet inhibition and major adverse cardiovascular and cerebrovascular events, particularly for intermediate metabolizers.

The Clinical Pharmacogenetics Implementation Consortium Guideline for SLCO1B1, ABCG2, and CYP2C9 genotypes and Statin-Associated Musculoskeletal Symptoms

Rhonda M. Cooper-DeHoff et al.

Summary: Statins are commonly prescribed medications that reduce cholesterol and prevent cardiovascular disease. However, statin-associated musculoskeletal symptoms can affect patient adherence and long-term effectiveness. Genetic variations in SLCO1B1, ABCG2, and CYP2C9 genes have been identified to impact the metabolism and adverse events of statins. This study summarizes the literature supporting these associations and provides therapeutic recommendations based on genotypes to improve the safety, adherence, and effectiveness of statin therapy.

Mayowa O. Owolabi et al.

Summary: Stroke is a major global threat to life and disability, particularly in low- and middle-income countries that struggle to address its challenges. This article provides an overview of primary stroke prevention, estimating the cost and identifying deficiencies in guidelines and primary prevention approaches. Pragmatic solutions are offered, emphasizing the role of governments and population-wide strategies. Implementation of primary stroke prevention involves various stakeholders across the lifespan.

Causal associations of alcohol consumption with cardiovascular diseases and all-cause mortality among Chinese males

Chunyu Hu et al.

Summary: This study finds a causal relationship between alcohol consumption and cardiovascular diseases (CVDs) and all-cause mortality in Chinese males. Light and moderate drinking can reduce the risk of CVDs and all-cause mortality. However, genetically predicted alcohol consumption shows a linear association with CVDs.

Racial and ethnic differences in pharmacotherapy to prevent coronary artery disease and thrombotic events

Juan Tamargo et al.

Summary: Awareness of racial/ethnic disparities in cardiovascular disease is crucial for providing effective healthcare. Differences in prevalence, risk factors, and drug response among different ethnic groups are mainly related to genetic variations. Understanding these differences helps optimize treatment and improve outcomes.

Stroke genetics informs drug discovery and risk prediction across ancestries

Aniket Mishra et al.

Summary: This study identified 89 independent loci associated with stroke and its subtypes through cross-ancestry GWAS analyses, with high replication rates for both primary and secondary stroke risk loci. It also revealed potential causal genes, variants, and drug targets, and demonstrated the predictive power of genetic risk scores for ischemic stroke across different ancestries.

Clinical Application of a Novel Genetic Risk Score for Ischemic Stroke in Patients With Cardiometabolic Disease

Nicholas A. Marston et al.

Summary: Genome-wide association studies have identified single-nucleotide polymorphisms associated with an increased risk of stroke. A genetic risk score (GRS) using 32 single-nucleotide polymorphisms was found to be a strong, independent predictor of ischemic stroke across a broad spectrum of subjects with cardiometabolic disease. In patients with atrial fibrillation and lower CHA(2)DS(2)-VASc scores, the GRS identified patients with a risk level comparable to those with higher scores.

Global, regional, and national burden of stroke and its risk factors, 1990-2019: a systematic analysis for the Global Burden of Disease Study 2019

Valery L. Feigin et al.

Summary: Regularly updated data on stroke including incidence, prevalence, mortality, disability, risk factors, and trends are crucial for evidence-based care planning. The Global Burden of Diseases Study provides comprehensive measurement of these metrics. From 1990 to 2019, while age-standardized rates improved, the number of strokes and deaths increased. Low-income countries had the highest stroke-related mortality and DALY rates, with high body-mass index being the fastest-growing risk factor.

Summary: A polygenic risk score was constructed and validated in a large Chinese population, showing individuals with high polygenic risk had a significantly increased risk of stroke, especially when combined with family history. Maintaining ideal cardiovascular health could mitigate the risk associated with high polygenic risk.
