Subretinal deposits in young patients treated with voretigene neparvovec-rzyl for RPE65-mediated retinal dystrophy

Jennifer Lopez et al.

Summary: This study reports the occurrence of subretinal deposits in three young patients following gene therapy for retinal dystrophy. However, these deposits improved or resolved over the follow-up period. These findings can provide insights into the delivery parameters and safety profile of future retinal gene therapy trials.

Summary: In this study, Voretigene Neparvovec was used to treat seven eyes of five patients with bi-allelic RPE65 mutations, showing improvements in rod function in most patients at 1 month. Visual acuity either slightly improved or remained stable, with cone function improvement in three eyes. Correlations were found between age and certain functional changes, suggesting potential parameters for therapy success.

Summary: This study investigates the safety and efficacy of voretigene neparvovec gene therapy in pediatric patients with biallelic RPE65 disease-causing variants. Results show significant improvements in visual function and retinal structure, including improved visual field and visual acuity, as well as small but statistically significant changes in central subfield thickness. Postoperative complications were generally manageable and resolved over time.

Summary: This study aimed to investigate the impact of patients' age, baseline visual acuity, and intraoperative foveal detachment on the outcomes of subretinal voretigene neparvovec-rzyl therapy, as well as the patients' perceptions of the treatment effect. The results showed that there were no significant vision changes for adult patients, while there was a trend of improvement for pediatric patients. The decrease in foveal thickness was observed, but its long-term significance remains to be determined.
