The Molecular Signature of Human Testicular Peritubular Cells Revealed by Single-Cell Analysis

N6-methyladenosine RNA demethylase ALKBH5 is testis-specifically downregulated in hybrid male sterile dzo and is a target gene of bta-miR-200a

The results show that the expression of m(6)A erasing, writing, and reading genes is significantly downregulated in dzo testis, while the expression of spermatogenesis-related genes is deregulated. Additionally, the promoter regions of FTO and YTHDC2 genes are highly methylated in dzo testis, and the expression of bta-miR-200a is downregulated, which targets m(6)A-associated genes. These findings reveal the important role of m(6)A-associated genes in dzo male sterility and suggest further exploration of the interaction between DNA methylation, miRNA, and m(6)A-associated protein expression.
