The potential anti-osteoporotic effect of exercise-induced increased preptin level in ovariectomized rats

Khaled Abdelfattah Abulfadle et al.

Summary: This study investigated the effects of estradiol administration and moderate exercise training on osteoporotic changes in ovariectomized rats. The results showed that both estradiol therapy and moderate exercise training significantly improved bone resorption and formation biomarkers, reduced pro-inflammatory cytokines, and increased bone osteopontin expression. These findings suggest that combined estradiol therapy and moderate exercise training may be a promising therapeutic approach for reducing postmenopausal osteoporosis.

Characterization of the metabolic differences between male and female C57BL/6 mice

Gabriel O. de Souza et al.

Summary: This study aimed to compare the responses of male and female C57BL/6 mice to various metabolic challenges. The results showed that male mice had stronger responses to food restriction and refeeding, while female mice had higher protection against diet-induced obesity. Additionally, male mice had different feeding responses to ghrelin and leptin compared to females. However, these sex differences were not explained by differences in central responsiveness or neuron fiber density.

Genetic ablation of the preptin-coding portion of Igf2 impairs pancreatic function in female mice

E. J. Buckels et al.

Summary: This study describes a mouse model in which the preptin-coding portion of the Igf2 gene has been genetically ablated. The mice do not exhibit reduced size at birth associated with Igf2 knockout, suggesting that IGF2 functionality is maintained. However, there is a change in the processing of mature Igf2. Female knockout mice show diminished glucose-stimulated insulin secretion, while the insulin response in males is not different from wild type.

Functional stapled fragments of human preptin of minimised length

Marta Lubos et al.

Summary: This study describes the synthesis, structures, and biological activities of six shortened analogues of human preptin. Stapled peptides with stabilized secondary structures were created by modifying the C-terminal part of preptin. The effect of these structural changes on calcium ion release was determined, and one of the analogues showed similar potency to the full-length preptin. These findings suggest the potential for designing new preptin analogues for diabetes and osteoporosis treatment.

The Impact of Maternal High-Fat Diet on Bone Microarchitecture in Offspring

Emma J. Buckels et al.

Summary: Research suggests that maternal high-fat diet may negatively impact embryonic bone development and bone volume in adult animals. The mechanisms behind these effects remain unclear, but some studies indicate sex-specific outcomes. Limited human studies also suggest that high-fat diet during pregnancy may impair bone formation and increase fracture risk in children.

Phenotypic sexual dimorphism in response to dietary fat manipulation in C57BL/6J mice

Isabel Casimiro et al.

Summary: The study revealed significant differences in responses to high-fat diet between male and female mice, particularly in body weight gain, food intake, energy expenditure, and insulin sensitivity. Male mice gained weight faster, had an increase in pancreatic beta cell area, and reduced insulin sensitivity after HFD feeding, while female mice did not exhibit these phenomena.

Summary: Bone fragility and obesity are multifactorial diseases with controversial impacts of high fat diet and overconsumption on bone health. Exercise and diet composition have different influences on bone characteristics, pointing to the complexity of the relationship between diet, obesity, and bone quality.

Hyperinsulinemia and Its Pivotal Role in Aging, Obesity, Type 2 Diabetes, Cardiovascular Disease and Cancer

Joseph A. M. J. L. Janssen

Summary: Hyperinsulinemia may be a significant factor in causing obesity, metabolic syndrome, cardiovascular disease, cancer, and premature mortality. Strategies for managing hyperinsulinemia include reducing insulin secretion, increasing hepatic insulin clearance, and maximizing insulin sensitivity.
