Drug-Repurposing Screening Identifies a Gallic Acid Binding Site on SARS-CoV-2 Non-structural Protein 7

Protein-protein interaction of RdRp with its co-factor NSP8 and NSP7 to decipher the interface hotspot residues for drug targeting: A comparison between SARS-CoV-2 and SARS-CoV

Summary: In this study, the molecular mechanism of the interaction between RdRp (Non-Structural Protein, NSP12) and its co-factors NSP7 and NSP8 in SARS-CoV-2 and SARS-CoV was explored. The results showed that the binding free energy of the SARS-CoV-2 heterotetramer complex with its corresponding partner protein was significantly higher compared to SARS-CoV. Interface hotspot residues were predicted and most of them clustered together. The overall replication complex of SARS-CoV-2 was slightly more flexible compared to SARS-CoV, which may explain the higher rate of RNA replication in SARS-CoV-2. The findings of the study provide insights for designing small molecules or peptide/peptidomimetic ligands to disrupt the interaction at the protein-protein interface.

Repurposed Antiviral Drugs for Covid-19-Interim WHO Solidarity Trial Results
