Single Diastereomers of the Clinical Anticancer ProTide Agents NUC-1031 and NUC-3373 Preferentially Target Cancer Stem Cells In Vitro

Summary: The synthesis of diastereomers of clinically active ProTides, NUC-1031 and NUC-3373, was described. The cytotoxic effects of these diastereomers were found to be preferentially targeting leukemic stem cells (LSCs), suggesting a potential mechanism involving accumulation of metabolized nucleoside analogues.
