Advances in human oxytocin measurement: challenges and proposed solutions

Benjamin A. Tabak et al.

Summary: Oxytocin, a neuropeptide known for its role in reproduction and socioemotional processes, may have therapeutic potential in treating social impairments, but the precise way to manipulate this system for clinical benefit is still unknown. The inconsistent use of standardized and validated oxytocin measurement methods, including the design and study of hormone secretion and biochemical assays, presents unresolved challenges.

Summary: Hypothalamic obesity in individuals treated for pediatric craniopharyngioma has a significant impact on social function. Families use various approaches for weight management, but medication trials are less common. Social function is more impaired in individuals with obesity.

A phase 3 randomized clinical trial using a once-weekly glucagon-like peptide-1 receptor agonist in adolescents and young adults with hypothalamic obesity

Christian L. Roth et al.

Summary: The study aimed to evaluate the efficacy and safety of GLP-1 RA in patients with HO. Although ExQW did not significantly change BMI after 36 weeks, it showed significant reductions in total body fat mass and waist circumference compared to placebo. GLP-1 RAs are a promising and safe treatment for improving or stabilizing HO in children and young adults.

Oxytocin in young children with Prader-Willi syndrome: Results of a randomized, double-blind, placebo-controlled, crossover trial investigating 3 months of oxytocin

Layla Damen et al.

Summary: The study on the effects of intranasal oxytocin treatment on children with Prader-Willi syndrome showed positive effects on social and eating behavior, especially in boys aged 3-11 years and in children with a deletion, with no safety concerns. However, individual effects should be carefully evaluated.

Summary: The study demonstrates that intranasal administration of OT can reliably increase endogenous salivary OT levels, particularly at the four-week follow-up after treatment cessation. Individuals with larger behavioral improvements in ASD social symptoms show more pronounced increases in salivary OT levels.

Oxytocin Administration Increases Proactive Control in Men with Overweight or Obesity: A Randomized, Double-Blind, Placebo-Controlled Crossover Study

Franziska Plessow et al.

Summary: The study explores the potential of oxytocin to improve cognitive control over behavior in humans. Findings suggest that oxytocin enhances proactive control over behavior by improving the ability to suppress behavioral impulses, which may have implications for its anorexigenic effects in human obesity. Future research is needed to further investigate the impact of oxytocin on cognitive control and its role in weight loss treatment.

Intranasal Oxytocin in Children and Adolescents with Autism Spectrum Disorder

L. Sikich et al.

Summary: A randomized trial involving 290 participants with autism spectrum disorder showed that daily use of intranasal oxytocin did not improve social interaction over a period of 24 weeks. Overall, there were no significant between-group differences in social or cognitive functioning between the oxytocin and placebo groups.

Oxytocin for Autism Spectrum Disorder - Down, but Not Out

Daniel H. Geschwind

Summary: Autism spectrum disorder is characterized by deficits in social functioning and repetitive-restrictive behaviors, with current pharmacologic interventions limited to symptom treatment instead of targeting social deficits. Behavioral treatments have shown effectiveness in some individuals, but there is a need for drug development to improve social functioning. The potential use of oxytocin as a pro-social hormone for autism treatment has gained attention.

Cerebral and extracerebral distribution of radioactivity associated with oxytocin in rabbits after intranasal administration: Comparison of TTA-121, a newly developed oxytocin formulation, with Syntocinon

Daisuke Ishii et al.

Summary: The study found that the newly developed TTA-121, compared to the traditional Syntocinon, increased the delivery of active substances in the whole brain by 3.6 times after single intranasal administration. Oxytocin may be transferred to the brain through various pathways such as the olfactory bulb and the trigeminal nerve. Results also showed an increase in radioactivity in the prefrontal area and the precuneus after intranasal administration, indicating possible sites of pharmacological action.

In the nose or on the tongue? Contrasting motivational effects of oral and intranasal oxytocin on arousal and reward during social processing

Juan Kou et al.

Summary: Oral administration of oxytocin increases arousal ratings for faces and associated brain reward responses, which is partially mediated by blood concentration changes. In contrast, intranasal administration of oxytocin decreases amygdala and arousal responses to face emotions. The route of administration of oxytocin can lead to significantly different effects on socioemotional cues and reward responses.

Hindbrain Administration of Oxytocin Reduces Food Intake, Weight Gain and Activates Catecholamine Neurons in the Hindbrain Nucleus of the Solitary Tract in Rats

Vishwanath T. Anekonda et al.

Summary: The study demonstrates the significant role of central nervous system oxytocin (OT) in regulating energy balance, particularly showing that OT in the hindbrain is effective at reducing food intake, weight gain, and adiposity. Additionally, it indicates that catecholamine neurons in the nucleus tractus solitarius (NTS) are downstream targets of CNS OT.

Effects of Combined Oxytocin and Beta-3 Receptor Agonist (CL 316243) Treatment on Body Weight and Adiposity in Male Diet-Induced Obese Rats

Melise M. Edwards et al.

Summary: The combined treatment of OT and the beta-3 agonist, CL 316243, produces an additive effect to decrease body weight and adiposity in DIO rats by reducing energy intake and increasing BAT thermogenesis. The effects were associated with decreased adiposity, adipocyte size, energy intake, and increased uncoupling protein 1 (UCP-1) content in epididymal white adipose tissue (EWAT), as well as elevated T-IBAT and IBAT UCP-1 content and IBAT thermogenic gene expression.
