Free radical as a double-edged sword in disease: Deriving strategic opportunities for nanotherapeutics

Lin Zhu et al.

Summary: This article reviews the double-edged roles of free radicals in biological systems and the application of nanotherapeutics in various disease treatments. It introduces the sources, types, and physiological functions of free radicals, focusing on nanotherapeutic approaches based on free radical generation and depletion. Specific emphasis is placed on the treatment of inflammatory diseases, neurodegenerative disorders, cardiovascular and cerebrovascular diseases, kidney/liver injury, diabetes and diabetic complications, tumorigenesis, etc. Ongoing challenges and perspectives of nanotherapeutic technologies based on regulating free radicals in biomedicine are discussed.

Chemodynamic Therapy via Fenton and Fenton-Like Nanomaterials: Strategies and Recent Advances

Chenyang Jia et al.

Summary: Chemodynamic therapy (CDT) is a novel cancer treatment strategy that generates hydroxyl radicals in the tumor region using Fenton or Fenton-like reactions. It has advantages such as low side effects, independence from external stimulation, modulation of the tumor microenvironment, and low treatment cost. CDT can also be combined with other therapies to enhance anticancer effects.

GSH-Depleted Nanozymes with Dual-Radicals Enzyme Activities for Tumor Synergic Therapy

Shengming Wu et al.

Summary: Developing a novel nanocatalyst platform involving longer half-life time ROS is desired to enhance tumor therapy efficacy. Fe3O4-Schwertmannite nanocomposites with triple-effect tumor therapy were successfully constructed, exhibiting remarkable tumor inhibition both in vitro and in vivo. The comprehensive properties of the nanoplatform, including dual-radical production, Fe3O4 nanocrystal mediated PTT, and the BSO mediated GSH depletion, pave a way to constructing a synergic catalytic nanoplatform for efficient tumor therapy.
