Engineering injectable, biocompatible, and highly elastic bioadhesive cryogels

Summary: We have recently developed biomimetic cryogel scaffolds that exhibit strong bioadhesive properties and can be syringe-injectable for tissue regeneration and remodeling. These cryogels, made from naturally-derived polymers and functionalized with mussel-inspired dopamine, were found to strongly adhere to various animal tissues and organs without cytotoxicity. In vivo data also showed good tissue integration and reduced inflammatory response. These biomimetic cryogels hold great promise for biomedical applications in wound healing, tissue engineering, and regenerative medicine.

Engineering hyaluronic acid-based cryogels for CD44-mediated breast tumor reconstruction

Summary: Breast cancer is a major health concern globally, especially among American women. Traditional therapies are usually ineffective and drug resistance often leads to cancer recurrence. To address this issue, the authors developed a method to generate uniform multi-cellular tumor spheroids and used cryogel scaffolds made from hyaluronic acid to reconstruct breast tumor microtissues with aspects of the tumor microenvironment. The resulting tumor model showed high aggressiveness and drug resistance, making it a valuable tool for preclinical research, drug screening, and cancer diagnosis.

Development of a letrozole-based synchronization protocol for fixed-time artificial insemination in beef cattle

Summary: This study aimed to determine the effectiveness of ovulation synchronization using a letrozole treatment of around 4 days (Exp 1), compare the efficacy of letrozole-based protocols with other commonly used synchronization protocols for fixed-time artificial insemination (FTAI) (Exp 2), and test a new intravaginal letrozole-releasing device (LRD) with and without pre-synchronization (Exp 3). The results showed that using letrozole for 4 days resulted in better ovulation synchrony in cows compared to other groups. The letrozole-based protocol had a lower pregnancy rate compared to other protocols in Exp 2. The new X-LRD device enhanced drug delivery compared to the T-LRD device in Exp 3.

The nanoparticles exhibited small size, narrow size distribution, high negative surface charge, and a biphasic sustained release of the extract. The FOL/TPP-decorated nanoparticles showed higher drug loading, enhanced toxicity to cancer cells, and improved cellular uptake compared to FOL-decorated nanoparticles. The FOL/TPP-NPs also demonstrated better stability, uptake efficiency, and targeting ability to mitochondria.

The nanoparticle enhanced drug delivery and accumulation in lymph nodes and tumors, and polarized M2 to M1 macrophages, resulting in immune microenvironment remodeling and preventing T cell exhaustion.

This approach enhances T cell-mediated secretion of cytotoxic proteins, restoring the killing ability of tumor-specific cytotoxic T lymphocytes both in vitro and in vivo, providing a safe platform to improve cancer immunotherapies.
