An Injectable Engineered Cartilage Gel Improves Intervertebral Disc Repair in a Rat Nucleotomy Model

Basanta Bhujel et al.

Summary: This study investigated the effectiveness of an engineered cartilage gel utilizing human fetal cartilage-derived progenitor cells (hFCPCs) on intervertebral disc repair in a rat tail nucleotomy model. The results showed that implantation of the cartilage gel promoted the repair of degenerative discs by increasing cellularity and matrix integrity, promoting reconstruction of nucleus pulposus, restoring disc hydration, and downregulating inflammatory cytokines and pain. These findings suggest that cartilage gel has greater therapeutic potential compared to cellular or ECM components alone.

Elsa Gonzalez-Cubero et al.

Summary: This study evaluates the therapeutic potential of mesenchymal stem cells secretome (CM) in the treatment of discogenic pain. The results show that compared to extracellular vesicles (EVs) and soluble culture fraction (SF), the presence of whole CM has a greater positive effect on regulating inflammation and inhibiting degradation factors. This suggests that CM may protect against degenerative disc disease and associated pain.

Substrate Topography Regulates Differentiation of Annulus Fibrosus-Derived Stem Cells via CAV1-YAP-Mediated

Genglei Chu et al.

Summary: In this study, fibrous poly(ether carbonate urethane)urea (PECUU) scaffolds with various fiber sizes were fabricated to mimic the microstructural feature of native annulus fibrosus (AF) and control the differentiation of AF-derived stem cells (AFSCs) by regulating topographical cues. The research demonstrated that the topography-dependent gene expression in AFSCs approximated the biochemical profile of AF tissue in different zones, indicating that tuning the fiber size of scaffolds can effectively induce changes in cell shape, adhesions, and extracellular matrix expression. The study may offer new insights for designing novel materials for AF tissue regeneration based on the activation of the CAV1-YAP mechanotransduction axis.
