Characterization of a Stemness-Optimized Purification Method for Human Dental-Pulp Stem Cells: An Approach to Standardization

Martin Philipp Dieterle et al.

Summary: This study analyzes human dental pulp stem cells (hDPSCs) using different purification methods and finds that STRO-1-positive cell enrichment is the optimal choice for maintaining stemness. Additionally, (immuno)phenotypic characterization is the minimum requirement for quality control in hDPSCs studies.

Soft substrates direct stem cell differentiation into the chondrogenic lineage without the use of growth factors

Tosca Roncada et al.

Summary: Mesenchymal stem cells (MSCs) are a promising treatment for cartilage injuries, but selectively promoting their differentiation in vivo is challenging. This study demonstrates for the first time that stiffness can guide MSC differentiation without the use of growth factors, using collagen and alginate scaffolds. This finding is important for developing tissue engineering scaffolds for the treatment of cartilage defects.

BMP4-regulated human dental pulp stromal cells promote pulp-like tissue regeneration in a decellularized dental pulp matrix scaffold

Qin Tan et al.

Summary: The study investigates the potential of decellularized dental pulp extracellular matrix (dECM) combined with bone morphogenetic protein 4 (BMP4) in regulating dental pulp stromal cells (DPSC) for pulp regeneration. The results show that dECM promotes DPSC proliferation, and the combination of BMP4 and dECM accelerates the formation of pulp-like tissue by DPSCs in vitro.

Summary: The periodontium is constantly exposed to mechanical forces, leading to cellular mechanotransduction processes that play a critical role in periodontal integrity and regeneration. Dysregulation of these processes can contribute to diseases such as periodontitis or oral squamous cell carcinoma. Future applications of mechanobiological principles in oral regenerative medicine show promise for addressing periodontal tissue diseases.
