Emerging skeletal muscle stromal cell diversity: Functional divergence in fibro/adipogenic progenitor and mural cell populations

Morten Ritso et al.

Summary: Healthy skeletal muscle relies on both skeletal muscle stem cells and tissue-resident stromal cells for repair and maintenance. Different subpopulations of stromal cells play distinct roles in the regenerative process, and dysregulation of communication among cell populations can lead to pathological changes and decline in muscle function. Restoring proper stromal cell function is crucial for therapeutic targeting and maintaining muscle homeostasis.

Interplay between mitochondrial reactive oxygen species, oxidative stress and hypoxic adaptation in facioscapulohumeral muscular dystrophy: Metabolic stress as potential therapeutic target

Philipp Heher et al.

Summary: FSHD is characterized by oxidative stress induced by DUX4, leading to metabolic dysfunction and impaired mitochondrial function. Increased mitochondrial ROS levels in FSHD muscle cells are associated with elevated steady-state mitochondrial membrane potential. DUX4 triggers mitochondrial membrane polarization, resulting in mitochondrial ROS generation and apoptosis.

Facioscapulohumeral muscular dystrophy-Reproductive counseling, pregnancy, and delivery in a complex multigenetic disease

Sanne C. C. Vincenten et al.

Summary: Reproductive counseling in FSHD can be challenging due to its complex genetic mechanisms and incomplete penetrance. Understanding the genetic causes and inheritance patterns is essential, especially with prenatal and preimplantation genetic diagnosis options now available for FSHD1. Clinicians should be aware of potential risks during pregnancy and delivery.

Summary: This minireview summarizes the current state of research in DUX4 and FSHD biology, highlighting key areas for further investigation to better understand DUX4 regulation and FSHD pathogenesis. The review also emphasizes post-transcriptional regulation of DUX4 through changes in RNA and protein stability, which may underlie key features of FSHD pathophysiology.

Persistent Fibroadipogenic Progenitor Expansion Following Transient DUX4 Expression Provokes a Profibrotic State in a Mouse Model for FSHD

Darko Bosnakovski et al.

Summary: FSHD is a disease caused by loss of silencing of the DUX4 gene, but the presence of the DUX4 protein in affected muscle has not been directly detected. This study used an animal model to investigate the extent of muscle regeneration following DUX4-mediated degeneration. The results showed that muscle histology could recover significantly after DUX4 expression was switched off, but the fibroadipogenic progenitor compartment remained elevated and the recovered muscle had a propensity for severe fibrosis. These findings have potential implications for therapeutic approaches.

Epigenetic regulation of satellite cell fate during skeletal muscle regeneration

Jimmy Massenet et al.

Summary: Muscle stem cells respond to muscle injury by integrating environmental cues to mediate regeneration, with changes in gene expression facilitated by cell signaling from the muscle environment. Age and disease alter the muscle environment, impacting the efficiency of cell fate transitions required for muscle repair, but the reversible nature of epigenetic changes offers potential for improving muscle regeneration in myopathies.

Muscle repair after physiological damage relies on nuclear migration for cellular reconstruction

William Roman et al.

Summary: The study revealed that localized muscle injuries triggered by exercise activate a myofiber self-repair mechanism that is independent of satellite cells. A signaling cascade involving calcium, Cdc42, and phosphokinase C attracts myonuclei to the damaged site, accelerating sarcomere repair and delivering messenger RNA for cellular reconstruction. Myofiber self-repair is a cell-autonomous protective mechanism, offering an alternative model for understanding muscle architecture restoration in health and disease.

DUX4 Role in Normal Physiology and in FSHD Muscular Dystrophy

Emanuele Mocciaro et al.

Summary: DUX4, a sequence-specific transcription factor, has evolved from obscurity to being recognized as a key player in physiological and pathological processes. Its highly regulated expression during embryonic development can lead to diseases such as FSHD, cancer, and viral infections through aberrant reactivation. Understanding the regulation and function of DUX4 could provide insights for therapeutic strategies targeting DUX4-associated diseases.

Pathomechanisms and biomarkers in facioscapulohumeral muscular dystrophy: roles of DUX4 and PAX7

Christopher R. S. Banerji et al.

Summary: Facioscapulohumeral muscular dystrophy (FSHD) is characterized by skeletal muscle weakness and wasting due to epigenetic derepression of the D4Z4 macrosatellite, leading to transcription of DUX4 which activates target genes. PAX7 suppression serves as a reliable biomarker for FSHD, but its link to genomic changes and DUX4 remains unclear. Understanding the roles of DUX4 and PAX7 in FSHD pathology can deepen knowledge of the disease through interactions with the immune system and muscle regeneration.

Summary: The research demonstrates that p53 activates Dux and DUX4 in mouse and human cells, promoting cell differentiation and ZGA. Mouse Dux is activated by p53 to drive differentiation in embryonic stem cells, and the p53-DUX/DUX4 regulatory axis is conserved in humans. This finding reveals potential therapeutic opportunities for FSHD.
