Synthesis of 2-aryl-3-triazolyl-indoles from phenacyltriazole-derived hydrazones: Exploring new scaffolds for anticonvulsant activity

Seyedeh Mahdieh Hashemi et al.

Summary: A series of novel 1-(2-(halophenyl)-2-(2-arylhydrazono)ethyl)-1H-1,2,4-triazoles 7a-l and their Fischer indolic products 2-(halophenyl)-3-(1H-1,2,4-triazol-1-yl)-1H-indoles 8a-l were synthesized conveniently. The (Z or E) geometry of arylhydrazones was identified by 1H NMR spectroscopy. The antiepileptic potential of both series of compounds was evaluated using PTZ and MES seizure models. The majority of hydrazones exhibited significant protective effects against MES-induced seizures, with compounds 7a and 7h being the most potent and rapid-acting. Indolic compounds generally displayed lower activity compared to their original arylhydrazones.

Evaluation of the increase of the thymoquinone permeability formulated in polymeric micelles: In vitro test and in vivo toxicity assessment in Zebrafish embryos

Jessika Lodovichi et al.

Summary: A polymeric micelles formulation (TQ-MP) using Soluplus(R)-Solutol(R) HS15 was developed to enhance the permeability of Thymoquinone (TQ) for possible oral and parenteral administration. The optimized micelles showed small size and homogeneous distribution, and in vitro experiments demonstrated improved permeability of TQ in the intestine and blood brain barrier.

Embryonic Arsenic Exposure Triggers Long-Term Behavioral Impairment with Metabolite Alterations in Zebrafish

Noraini Abu Bakar et al.

Summary: This study provides new insight into the potential mechanism of arsenic toxicity leading to long-term learning impairment in zebrafish. The researchers found that exposure to arsenic trioxide (As2O3) resulted in increased mortality and various behavioral deficits in zebrafish, including motor behavior deficits and impaired color preference. The study also identified changes in gene regulation and metabolic pathways associated with the toxic effects of As2O3. These findings may be useful for future risk assessments of environmental toxins.

Identification of curcumin analogues with anti-seizure potential in vivo using chemical and genetic zebrafish larva seizure models

Brandon Kar Meng Choo et al.

Summary: Seizures are abnormal brain activities that can be challenging to manage with currently available drugs. Curcumin, a compound with anti-seizure properties, has poor bioavailability. This study tested structural analogues of curcumin in zebrafish models and found promising candidates for novel anti-seizure agents.

Tail Coiling Assay in Zebrafish (Danio rerio) Embryos: Stage of Development, Promising Positive Control Candidates, and Selection of an Appropriate Organic Solvent for Screening of Developmental Neurotoxicity (DNT)

Andreia A. S. de Oliveira et al.

Summary: The tail coiling assay in zebrafish embryos is proposed as an alternative method for screening developmental neurotoxicity induced by chemicals. Embryos from 22 to 29 hpf showed consistent coiling activity, with caffeine and MS-222 affecting activity levels differently. Using 1% DMSO as a solvent for the assay was considered relatively safe.

Zebrafish as an animal model for biomedical research

Tae-Young Choi et al.

Summary: Zebrafish are emerging as a powerful animal model for studying mental disorders and metabolic diseases due to their transparency, low maintenance costs, and genetic similarity to humans. Researchers highlight the importance of zebrafish in understanding communication pathways between the brain and other organs, as well as the potential for tracking metabolism and disease progression. The ease of tissue imaging, high offspring numbers, and availability of genetic techniques further contribute to the benefits of using zebrafish in biomedical research.

Brain concentrations of glutamate and GABA in human epilepsy: A review

Gabrielle L. Sarlo et al.

Summary: The imbalance between excitation and inhibition in epilepsy is related to changes in glutamate and GABA levels, with most studies showing increased glutamate in epileptic regions. Future research should consider medication use, measure GABA, glutamate, and glutamine levels, use high-tesla strength MRI, and evaluate the timing of microdialysis further.

Zebrafish Bioassay for Screening Therapeutic Candidates Based on Melanotrophic Activity

Ted Hong et al.

Summary: This study established a bioassay using the zebrafish animal model to measure the physiological efficacy differential of alpha-MSH analogues. The results showed that unique alpha-MSH derivatives from several fish species produced differential effects on melanophore dispersion, with the synthetic analogue melanotan-II exhibiting higher physiological effects. The findings suggest that the melanophore assay may serve as a useful screening tool for therapeutic candidates in drug discovery.

Mohanapriya Gunasekharan et al.

Summary: Bacterial infections, especially MRSA, are a major cause of morbidity and mortality. New non-PAINS associated pyrrolylated-chalcones were synthesized and evaluated for their potency against MRSA, with hydroxyl-containing compounds showing the most significant anti-MRSA efficiency.
