Understanding interactions among flavor compounds from spices and myofibrillar proteins by multi-spectroscopy and molecular docking simulation

Summary: This study investigated the influence of constant heating treatment on the structural and adsorption properties of chicken myofibrillar proteins (MPs). The results showed that heat treatment increased the exposure of sulfhydryl groups and improved the hydrophobicity of MPs' surface. The particle size distribution, alpha-helix and beta-sheet structures, as well as the formation of aggregates, viscosity, surface roughness, and zeta potential of MPs were significantly affected by heat treatments. Additionally, the binding interactions between MPs and flavors of spices varied depending on the nature of MPs, flavor compounds, and heat treatment duration.

Insight into the protein oxidation impact on the surface properties of myofibrillar proteins from bighead carp

Summary: The influence of oxidative modifications of myofibrillar proteins (MPs) on their surface properties was investigated in this study. Oxidized MPs were measured for oxidative modifications (deamination, formation of disulfide bonds and Schiff bases), particle size, net surface charge, and binding ability of volatiles (2-enthylfuran, 1-octen-3-ol, hexanal, and octanal). Molecular docking of volatiles with actomyosin was performed, and specific oxidative modifications (monoxidation and deamination) of MPs were determined using LC-MS/MS. The results showed that oxidative modifications of MPs changed their surface properties, including enhanced surface hydrophobicity and decreased affinity to volatile compounds and water. This study provides evidence of how protein oxidation affects the properties of MPs and deteriorates fish meat quality.

Effect of sodium bicarbonate and sodium chloride on protein conformation and gel properties of pork myofibrillar protein

Summary: The study investigated the effects of using sodium bicarbonate instead of sodium chloride on the endogenous fluorescence, ultraviolet absorption spectrum, water holding capacity, gel properties, and water distribution of pork myofibrillar protein. With the increase in sodium bicarbonate concentration, the pH increased, resulting in more tryptophan and tyrosine residues being exposed, which significantly improved the cooking yield, gel strength, and reduced centrifugal loss of the protein. However, the color of the cooked protein became darker and the bound water and immobile water became more tightly bound to the protein matrix, except in the sample with 0.4% sodium bicarbonate. It was also found that the mobility of water was enhanced when 0.4% sodium bicarbonate was added.

Probing the interaction between selected furan derivatives and porcine myofibrillar proteins by spectroscopic and molecular docking approaches

Summary: The interactions between myofibrillar proteins (MPs) and furan derivatives were studied using spectroscopic and molecular docking approaches. The binding capacity of MPs to furan derivatives was found to be influenced by molecular polarity and the position of the branched chain. Fluorescence analysis revealed that the interaction involved both static and dynamic quenching, affecting the microenvironment of tryptophan and tyrosine residues in MPs. Conformational investigation showed that the presence of furan derivatives transformed the ordered structures of MPs into random coils. Molecular docking and thermodynamic parameters indicated that hydrogen bonds, van der Waals forces, and hydrophobic interactions were the main interaction forces between MPs and the four furan derivatives.

Molecular dynamics simulation of the interactions between sesamol and myosin combined with spectroscopy and molecular docking studies

Summary: Spectroscopy and molecular dynamics were used to study the structural characteristics of Lacobacillus brevis R4 protease under different pH and temperature conditions. The protease showed stable spatial structure at pH 7 and 40 degrees C, while changes in the secondary structure were observed at pH 8 and 70 degrees C. Additionally, various vibrations and bonds were identified in the protease at different pH and temperatures. Molecular docking analysis suggested that the protease may interact with meat proteins to enhance meat tenderness and improve the quality of Harbin dry sausages.
