Metabolomics Combined with Transcriptomics Analysis Revealed the Amino Acids, Phenolic Acids, and Flavonol Derivatives Biosynthesis Network in Developing Rosa roxburghii Fruit

Summary: In this study, the metabolomics of Rosa roxburghii fruit at three developmental stages were analyzed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). A total of 533 metabolites were identified, with 339 significantly altered. The correlation between total phenols, flavonoids, amino acids and in vitro antioxidant activity was determined. Furthermore, the co-enrichment analysis of metabolome and transcriptome focused on amino acid, phenylpropanoid, and flavonoid biosynthesis pathways. The networks of amino acid, phenolic acid, and flavonol biosynthesis were constructed, and the gene expression was detected in different R. roxburghii genotypes. Key metabolites were identified in the in vitro antioxidant activities at different developmental stages and in different genotypes of R. roxburghii. This study provides novel insights into the synthesis of flavonoids, polyphenols, and amino acids in R. roxburghii.

Summary: The aim of this study was to investigate the liberation and phytochemical profile of bound polyphenols present in dietary fiber isolated from Rosa roxburghii fruit pomace, and to explore their biological activity and prebiotic potential. The results showed that the bound polyphenols were released during colonic fermentation at a higher ratio compared to gastrointestinal digestion, and they exhibited antioxidant and alpha-glucosidase inhibitory activity. Furthermore, the study demonstrated that the dietary fiber had a stronger prebiotic effect, promoting the growth of beneficial bacteria and the production of short chain fatty acids. The bound polyphenols may serve as important functional food ingredients in Rosa roxburghii fruit pomace.
