Enhanced Cancer Starvation Therapy Enabled by an Autophagy Inhibitors-Encapsulated Biomimetic ZIF-8 Nanodrug: Disrupting and Harnessing Dual Pro-Survival Autophagic Responses

Fenglan Li et al.

Summary: In this study, a novel nanodrug system was designed to achieve an enhanced anticancer effect through an autophagy regulation strategy and starvation therapy. The results showed that ZIF-8 as a nanocarrier could induce autophagy for promoting cancer cell survival, while CQ could inhibit this autophagy and achieve the anticancer effect. Moreover, starvation caused by GOx could harness autophagy for maintaining intracellular ATP levels, and CQ could further inhibit autophagy and enhance the anticancer efficiency of starvation therapy.

C-reactive protein perturbs alveolar bone homeostasis: An experimental study of periodontitis and diabetes in the rat

Mengjiao Zhou et al.

Summary: This study explores the role of C-reactive protein (CRP) in periodontitis and diabetes, as well as its mechanism in alveolar bone homeostasis. The results show that in periodontitis and diabetes, CRP can inhibit osteogenesis and promote osteoclastogenesis through the PI3K/AKT signaling pathway, thus disrupting alveolar bone homeostasis.

Yuxiao Feng et al.

Summary: This study reports a new hierarchical micro-mesoporous zeolitic imidazolate framework-8 (ZIF-8) with core-shell superstructure (HZIF-8) for enzyme immobilization. The immobilized enzyme in HZIF-8 exhibited higher activity and substrate affinity compared to conventional microporous ZIF-8, showing improved stability and reusability. The obtained HZIF-8 also showed outstanding performance in co-immobilization of multienzyme for glucose detection.

Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Control of gasdermin D oligomerization and pyroptosis by the Ragulator-Rag-mTORC1 pathway

Charles L. Evavold et al.

Summary: The Ragulator-Rag complex is found to be necessary for GSDMD pore formation and pyroptosis in macrophages, promoting GSDMD oligomerization on the plasma membrane. Defects in GSDMD oligomerization and pore formation can be rescued by mitochondrial poisons that stimulate ROS production, and ROS modulation impacts the ability of inflammasome pathways to promote pore formation downstream of GSDMD cleavage.
