Summary: In this study, a pioneering grafting from drug coordination polymerization strategy was proposed to develop a fully-coordinated and full-active Cu2O/drug core/shell nanoparticles for multimodal cancer therapy. The nanoplatform exhibited a rapid acid-responsive dissociation behavior and released chemotherapeutic drug cisplatin (CDDP) and photosensitizer indocyanine green (ICG) to achieve efficient triple combined photodynamic therapy/chemodynamic therapy/chemotherapy. The combination of the nanoplatform and alpha PD-1 effectively inhibited distal tumor growth, lung metastasis, and tumor recurrence through reprogramming tumor microenvironment and activating CD8+ T cell antitumor immunity. This study provides a new direction for the design of nanodrug delivery systems.

Polyphenol-Based Nanomedicine Evokes Immune Activation for Combination Cancer Treatment

The nanocomplex can induce chemotherapeutic/photodynamic immunogenic cancer cell death upon laser irradiation, supported by dendritic cell maturation, secretion of inflammatory cytokines, and inhibition of distant untreated tumors.
