Xiaojun Zhou et al.

Summary: The study demonstrates the construction of a dual-factor delivery scaffold, which combines short nanofibers and a 3D printed scaffold, for guiding vascularized bone regeneration. The scaffold exhibits an adjustable porous structure and strong compressive strength. The sequential release of the loaded factors promotes angiogenesis and osteogenesis, accelerating tissue ingrowth and vascularized bone regeneration. This study provides a promising strategy for constructing a bone microenvironment-matched biomimetic scaffold for bone regeneration.

Fabrication and evaluation of a BMP-2/dexamethasone co-loaded gelatin sponge scaffold for rapid bone regeneration

Qi Gan et al.

Summary: This study evaluated the osteogenic activity of dual drugs (Dex and BMP-2) loaded into a gelatin scaffold in vivo, and investigated the mechanism of osteogenesis induced by these drugs using C2C12 cells. Results showed that the gelatin scaffold with Dex and BMP-2 significantly accelerated osteogenesis in vivo, and Dex enhanced the osteogenic effect of BMP-2 through activation of the Runx2 pathway.

Environmentally friendly fabrication of electrospun nanofibers made of polycaprolactone, chitosan and k-carrageenan (PCL/CS/k-C)

Zulema Vargas-Osorio et al.

Summary: Electrospun nanofibers based on biodegradable polymers were successfully fabricated, and it was found that the addition of k-carrageenan improved the fabrication process. After one month of storage, the properties of the fiber mats were not significantly compromised, indicating a reduction in waste production. Cell viability assays confirmed that all prepared fiber mats promoted cell proliferation, indicating their potential applications in bone tissue engineering and wound healing.

Sustained release of BMP-2 using self-assembled layer-by-layer film-coated implants enhances bone regeneration over burst release

MayLin T. Howard et al.

Summary: This study investigated the design of different laponite clay diffusional barrier layer architectures in self-assembled LbL films to modulate the release kinetics of bone morphogenetic protein 2 (BMP-2) for bone regeneration. The results showed that sustained release of BMP-2 over 30 days significantly enhanced bone regeneration, while release over 2 days did not promote additional bone growth.

Graphene Hydrogel as a Porous Scaffold for Cartilage Regeneration

Chengqi Lyu et al.

Summary: Porous scaffolds are commonly used for cartilage tissue regeneration, but understanding the impact of their hierarchical structure on the regeneration process can be challenging. In this study, a graphene hydrogel with stable and adjustable structure was used as a model scaffold to investigate the effect of porous structure on matrix remodeling and chondrocyte ingrowth. The results showed that the graphene scaffold with an open pore structure accelerated and balanced cartilage remodeling. Furthermore, this enhanced remodeling selectively promoted the expression of collagen type II fibrils, providing new insights into scaffold design for cartilage repair.

Strontium-incorporated bioceramic scaffolds for enhanced osteoporosis bone regeneration

Qianju Wu et al.

Summary: This study aimed to fabricate bioactive scaffolds by incorporating strontium into modified amino-functional mesoporous bioactive glass (MBG) and investigate their bioactivity in vitro and in vivo. The results showed that the strontium-incorporated scaffolds had good biocompatibility and enhanced osteogenic and angiogenic capacities. In addition, bioinformatics analysis suggested that the scaffolds could reduce reactive oxygen species levels in bone marrow mesenchymal stem cells in the osteoporotic model, playing an anti-osteoporosis role while promoting osteogenesis.
