Comparative efficacy and safety of biologic agents in patients with active rheumatoid arthritis and inadequate response to tumor necrosis factor inhibitors: A Bayesian network meta-analysis of randomized controlled trials

Yoon-Kyoung Sung et al.

Summary: A meta-analysis using Bayesian network was conducted to evaluate the efficacy and safety of biologic agents in patients with rheumatoid arthritis who show inadequate response to tumor necrosis factor inhibitors. The results showed that abatacept, rituximab, and tocilizumab appeared to be more efficient in treating TNF inhibitor-refractory RA patients compared to other biologics, with no significant difference in safety.

A tumor-specific pro-IL-12 activates preexisting cytotoxic T cells to control established tumors

Diyuan Xue et al.

Summary: A tumor-conditional IL-12, pro-IL-12, was developed to selectively activate TILs and control tumor growth with reduced toxicity. Pro-IL-12 improved the efficacy of immune checkpoint blockade and targeted therapy when used in combination.

Exploiting protease activation for therapy

Chloe Bleuez et al.

Summary: This article provides an overview of therapeutic approaches based on protease activation and highlights recent insights into the spatial and temporal control of proteolytic activation. Two emerging areas of research are covered: antibody therapies targeting the proteolytic activation process of protein precursors and the use of engineered biopharmaceuticals with proteolytically activable pro-forms to increase specificity and improve therapeutic effectiveness.

The Uniqueness of Albumin as a Carrier in Nanodrug Delivery

Alessandra Spada et al.

Summary: Albumin is an attractive carrier in nanomedicine due to its high biocompatibility, ability to interact with various drugs, and active targeting potential. It has a long serum half-life of around 19 days, promoting half-life extension and targeted drug delivery. The article emphasizes the importance of albumin as a drug delivery system and highlights its potential future trends in drug delivery.

A Tale of Two Immune Cells in Rheumatoid Arthritis: The Crosstalk Between Macrophages and T Cells in the Synovium

Jiajie Tu et al.

Summary: RA is a chronic inflammatory autoimmune disease characterized by joint inflammation, immune cell infiltration, synovial hyperplasia, and excessive production of proinflammatory cytokines. The interaction between monocytes/macrophages and T cells in the joint synovium of RA patients plays a crucial role in promoting the development of RA. Further understanding of these interactions may lead to new perspectives on RA pathogenesis and targeted therapy development.
