The Impact of Systemic Inflammation on Alzheimer's Disease Pathology

Junhua Xie et al.

Summary: Alzheimer's disease is an age-related neurodegenerative disorder with increasing prevalence. Current treatments are limited to symptomatic medication, and there is a need for a better understanding of the underlying mechanisms to develop more rational therapies. Peripheral inflammation has been identified as a detrimental factor in AD, and this review provides an overview of the link between peripheral inflammation and AD initiation and progression, contributing to a broader understanding of the disease pathology.

Inflammatory bowel disease is associated with higher dementia risk: a nationwide longitudinal study

Bing Zhang et al.

Summary: The study found a significant association between IBD and the development of dementia, with IBD patients having a higher risk of dementia at an earlier age. The risk of developing Alzheimer's dementia was particularly increased among IBD patients. Research is needed to further explore the relationship between IBD and dementia.

Gut Inflammation Induced by Dextran Sulfate Sodium Exacerbates Amyloid-β Plaque Deposition in the AppNL-G-F Mouse Model of Alzheimer's Disease

Mona Sohrabi et al.

Summary: The study investigated the impact of chronic intestinal inflammation on brain changes in Alzheimer's disease (AD) using a mouse model. The results demonstrated that increased gut inflammation altered plaque deposition and glial immunoreactivity in the brain of AD model mice. This study contributes to our understanding of the influence of peripheral inflammation on A beta deposition via an inflammatory bowel disease-like model system.

Fibrin is a critical regulator of neutrophil effector function at the oral mucosal barrier

Lakmali M. Silva et al.

Summary: Tissue-specific cues, particularly fibrin, play a critical role in regulating neutrophil function at mucosal barriers, with commensal microbiota triggering extravascular fibrin deposition. Fibrin engages neutrophils through integrin receptors and activates immune-protective functions, but impaired fibrinolysis can lead to tissue damage. Polymorphisms in plasminogen gene are associated with periodontal disease, suggesting that fibrin-neutrophil engagement may contribute to mucosal diseases.

The neutrotime transcriptional signature defines a single continuum of neutrophils across biological compartments

Ricardo Grieshaber-Bouyer et al.

Summary: Neutrophils exhibit a single developmental spectrum, ranging from immature pre-neutrophils in the bone marrow to mature neutrophils in the blood and spleen. Inflammation affects neutrophil transcriptional states, leading to varying transcriptional activities at different sites.

Skull and vertebral bone marrow are myeloid cell reservoirs for the meninges and CNS parenchyma

Andrea Cugurra et al.

Summary: The meninges contain a pool of monocytes and neutrophils supplied by adjacent skull and vertebral bone marrow, rather than by the blood. Under pathological conditions, CNS-infiltrating myeloid cells can originate from the brain borders and have distinct transcriptional signatures from their blood-derived counterparts. Therefore, myeloid cells from adjacent bone marrow niches are strategically placed to supply innate immune cells under both homeostatic and pathological conditions.

Heterogeneity of meningeal B cells reveals a lymphopoietic niche at the CNS borders

Simone Brioschi et al.

Summary: The meninges contain adaptive immune cells called B cells that are thought to originate locally from the calvaria, providing a constant supply of CNS-educated B cells. Additionally, a subset of antigen-experienced B cells in the meninges of aging mice are found to be blood-borne, suggesting a potential mechanism for maintaining immune privilege within the CNS.
