Suppression of Fli-1 protects against pericyte loss and cognitive deficits in Alzheimer's disease

Pengfei Li et al.

Summary: The transcription factor Fli-1 plays a role in the loss of pericytes, inflammatory response, AD deposition, vascular dysfunction, and cognitive decline in AD. Inhibition of Fli-1 may represent a novel therapeutic strategy for AD.

Role of the CXCL13/CXCR5 Axis in Autoimmune Diseases

Zijian Pan et al.

Summary: CXCL13 and its receptor CXCR5 play an important role in the pathogenesis of autoimmune diseases. This review discusses the biological features of CXCL13 and CXCR5, and the recent findings on their pathogenic roles in autoimmune diseases. Furthermore, the potential use of CXCL13 as a disease biomarker and therapeutic target in autoimmune diseases is also discussed.

Expression of GM-CSF Is Regulated by Fli-1 Transcription Factor, a Potential Drug Target

Xuan Wang et al.

Summary: This study demonstrated that Fli-1 regulates the expression of GM-CSF in both T cells and endothelial cells through direct binding to the GM-CSF promoter and phosphorylation. Fli-1 also acts in an additive manner with another transcription factor Spl to regulate GM-CSF expression. Additionally, a low dose of the chemotherapeutic drug camptothecin inhibited the expression of Fli-1 and reduced GM-CSF production in human T cells. These findings provide insights into novel mechanisms for modulating GM-CSF expression and highlight Fli-1 as a critical regulator of inflammation and immunity that can be targeted for therapeutic intervention.

Camptothecin and Topotecan, Inhibitors of Transcription Factor Fli-1 and Topoisomerase, Markedly Ameliorate Lupus Nephritis in (NZB x NZW)F1 Mice and Reduce the Production of Inflammatory Mediators in Human Renal Cells

Xuan Wang et al.

Summary: The study showed that low-dose camptothecin (CPT) and topotecan (TPT) could be repurposed to treat lupus nephritis by reducing proteinuria levels, attenuating renal injury, decreasing autoantibody levels, and reducing deposits in the glomeruli.

CXCL13 Neutralization Attenuates Neuropsychiatric Manifestations in Lupus-Prone Mice

Michelle W. Huang et al.

Summary: Treatment with anti-CXCL13 significantly improves cognitive function and depression-like behavior in MRL/lpr mice, regardless of the route of administration. However, it does not substantially change the extent of lymphocyte infiltration into the brain or the general composition of the aggregates. Analysis of brain gene expression in anti-CXCL13 treated mice reveals significant differences in key immunological and neuro-inflammatory pathways, which likely explain the improvement in behavioral phenotype.
