Characterization of a Bioink Combining Extracellular Matrix-like Hydrogel with Osteosarcoma Cells: Preliminary Results

Design and biofabrication of bacterial living materials with robust and multiplexed biosensing capabilities

Summary: The combination of synthetic biology and 3D bioprinting has the potential to improve various applications by producing engineered living materials (ELMs) with genetically-encoded sense and response capabilities. Efforts are still needed to simplify the fabrication of sensing ELMs suitable for field use and enhance their functional complexity. In this study, bacterial ELMs with synthetic biosensing circuits were successfully constructed and demonstrated their performance in harsh conditions. Multiple biosensor strains were also incorporated into the ELMs to create materials with diverse sensing capabilities and cell-cell communication for pattern formation. This research paves the way for automating the prototyping process and expanding the applications of ELMs in monitoring and sensing.

Combined Analytical Approaches to Standardize and Characterize Biomaterials Formulations: Application to Chitosan-Gelatin Cross-Linked Hydrogels

Summary: A protocol combining different analytical methodologies was proposed to standardize the experimental conditions for reproducible formulations of hybrid hydrogels. The hybrid material, composed of gelatin and chitosan functionalized with methylfuran and cross-linked with 4-arm-PEG-maleimide, mimics the role, dynamics, and structural complexity of the extracellular matrix. Physical-chemical properties of the starting polymers and final constructs were characterized using a combination of HP-SEC-TDA, UV, FT-IR, NMR, and TGA.
