Trojan-Horse Diameter-Reducible Nanotheranostics for Macroscopic/Microscopic Imaging-Monitored Chemo-Antiangiogenic Therapy

Summary: This study presents a carrier-free Trojan-horse diameter-reducible metal-organic nanotheranostics that can enhance targeting efficiency, tumor penetration, and cellular uptake. This nanotheranostics is responsively decomposed into ultra-small-diameter nanotheranostics under stimuli of the tumor microenvironment, and it can be internalized into tumor cells through tumor-receptor-mediated self-targeting. Additionally, it enables visualization of self-targeting capacity and possesses an excellent antiangiogenic effect.

Additionally, the nanoparticles upregulate OX40 on intratumoral CD8+ T cells and synergize with an agonistic OX40 antibody to suppress tumor growth by 89.2%.

Summary: The release of PGE2 from dying cancer cells, mediated by COX-2, contributes to resistance against cytotoxic therapy. This study shows that cytotoxic drugs only induce PGE2 release in cancer cells with basal COX-2 activity, and that pharmacological COX-2 inhibition can enhance the efficacy of chemotherapy and PD-1 blockade combination. Additionally, all types of chemotherapy drugs enhance COX-2 transcription and inhibit cancer cell proliferation.

Synthesis of salicylic acid phenylethyl ester (SAPE) and its implication in immunomodulatory and anticancer roles

Summary: Salicylic acid phenylethyl ester (SAPE) was synthesized and studied for its immunomodulatory and anticancer properties. It exhibited low toxicity and favorable physical properties. Molecular docking showed better performance of SAPE compared to ibuprofen and indomethacin. In vitro experiments demonstrated potential efficacy of SAPE in hemolysis, tumor inhibition, and anti-inflammation. It also showed promising results in inhibiting breast cancer cell growth and inducing apoptosis. In a colitis-induced rat model, SAPE reduced disease activity index and tissue disruption, while increasing anti-oxidative enzymes and decreasing pro-inflammatory cytokines.
