Effects of aneuploidy on cell behaviour and function

Rong Li et al.

Summary: Aneuploidy, characterized by deviations in the copy number of chromosomes, affects organisms from early development through to aging and is a main cause of human pregnancy loss and cancer. Recent studies have increased our understanding of its mechanisms and how it can be both beneficial and detrimental, depending on the karyotype and external cues. These insights shed light on its roles in human pathogenesis and genome evolution.

Global profiling of phosphorylation-dependent changes in cysteine reactivity

Esther K. Kemper et al.

Summary: Proteomics has revealed the vast diversity of proteins, or proteoforms, generated from around 20,000 human genes, largely due to post-translational modifications (PTMs). This study introduces a chemical proteomic method to quantitatively study the impact of serine/threonine phosphorylation on the reactivity of cysteine residues. The results show that phosphorylation can significantly affect the reactivity of cysteine residues, highlighting the potential for creating small-molecule probes that target proteoforms with PTMs.

Aurora A-mediated pyruvate kinase M2 phosphorylation promotes biosynthesis with glycolytic metabolites and tumor cell cycle progression

Ya Jiang et al.

Summary: This study reveals a new protumor function of Aurora A by phosphorylating PKM2 during the S phase of the cell cycle to promote glycolysis and tumor growth. The spatial and temporal regulation of the specific kinase and PKM2 interaction is context dependent, showing intricate interconnectivity between cell cycle and glycolytic regulators.

G6PD: A hub for metabolic reprogramming and redox signaling in cancer

Hung-Chi Yang et al.

Summary: Metabolic hubs are crucial in cancer initiation and development. G6PD, a pivotal factor, affects cell proliferation and survival through altering redox homeostasis. The aberrant activation of G6PD via metabolic reprogramming can lead to either oxidative damage or oncogenic signaling in cancer cells.

Aneuploidy renders cancer cells vulnerable to mitotic checkpoint inhibition

Yael Cohen-Sharir et al.

Summary: The study shows that aneuploid cancer cells are more sensitive to genetic perturbation of SAC components and their long-term proliferation is jeopardized. Furthermore, aneuploid cells exhibit aberrant spindle geometry and dynamics, which result in mitotic defects and less-fit karyotypes over time.

Gene copy-number changes and chromosomal instability induced by aneuploidy confer resistance to chemotherapy

Summary: The study found that transient chromosomal instability can accelerate the development of resistance to anti-cancer therapies in tumor cells, as well as provide phenotypic plasticity for them to adapt to various stressful environments. Furthermore, aneuploidy may be an underexplored cause of therapy failure in human tumors.
