Advances, challenge and prospects in cell-mediated nanodrug delivery for cancer therapy: a review

Summary: Nanomedicine has great potential in improving drug effectiveness and reducing toxicity for tumor therapy. However, its application in clinical trials has been limited by various physiological barriers. Circulating cell-mediated nano-drug delivery systems offer a promising solution by collaborating with nano-drug delivery systems to overcome these barriers. Despite its advantages such as excellent biocompatibility and long-circulation time, there are still limitations to implementing this strategy.

A nanoplatform self-assembled by coordination delivers siRNA for lung cancer therapy

Summary: The high incidence and mortality of lung cancer pose a global threat. Clinical trials have shown that mutations in certain genes directly or indirectly affect the progress of lung cancer. As a result, molecular inhibitors have been found to benefit patients in clinical therapy, helping to prolong survival time and showing great potential in lung cancer treatment. siRNA, a nucleic acid molecule, can silence targeted gene translation by binding to mRNA. In this study, siRNA is delivered by coordination with Fe2+ to form a self-assembly structure, with doxorubicin (DOX) and human serum albumin (HSA) used for stabilization. The nanoplatform exhibited good lysosome escape, gene silencing effect, and cytotoxicity, providing new possibilities for co-delivery of siRNA and other molecular or protein drugs.

Iron Oxide Nanoparticle-Mediated siRNA Delivery System for Disease Treatment

Summary: Huntington's disease (HD) is a genetic disorder that mainly affects neurons in the striatum due to the presence of a toxic protein. Nanoparticles (NPs) have been developed as a promising delivery system for therapeutic RNA in treating HD. In this study, magnetic NPs (MNPs) were generated and characterized, and they showed low cytotoxicity and high transfection efficiency in delivering interfering RNA into cells. The MNPs were able to downregulate the production of the toxic protein, suggesting their potential as a treatment strategy for HD.

Zwitterionic Modification of Polyethyleneimine for Efficient In Vitro siRNA Delivery

Summary: Polyethylenimine (PEI) is commonly used for gene delivery, but its high cytotoxicity and non-specific protein adsorption limit its efficacy and practical applications. In this study, hydrophobically modified and zwitterion-modified PEIs were synthesized and demonstrated to be efficient gene carriers. These modified PEIs showed excellent siRNA binding capacity, reduced cytotoxicity, nonspecific protein adsorption, and enhanced stability. The findings suggest that zwitterionic modification is an effective method to improve the performance of polycationic gene carriers.

miR-34a-5p inhibits the malignant progression of KSHV-infected SH-SY5Y cells by targeting c-fos

Summary: miR-34a-5p was downregulated in KSHV-infected SK-RG cells. Restoration of miR-34a-5p led to decreased cell proliferation and migration, G1 cell cycle arrest, and downregulation of CDK4/6, cyclin D1, MMP2, MMP9. c-fos was identified as the target of miR-34a-5p, and upregulation of c-fos weakened the effects of miR-34a-5p. Xenograft tumor experiments and immunohistochemistry assays confirmed the inhibitory effects of miR-34a-5p on tumor growth and virus gene expression.

Synthetic Helical Polypeptide as a Gene Transfection Enhancer
