Vascular Endothelial Growth Factor as Molecular Target for Bronchopulmonary Dysplasia Prevention in Very Low Birth Weight Infants

Summary: Bronchopulmonary dysplasia (BPD) is still a significant burden in neonatal care, and its definition is currently based on treatment rather than diagnostic criteria. BPD is associated with various prenatal and postnatal risk factors, causing distorted lung development and function. Inflammation and oxidative stress play key roles in BPD development, with vascular endothelial growth factor-A (VEGFA) being an important candidate. This educational review discusses the inflammatory pathways in BPD and highlights the role of VEGFA, providing an overview of current evidence.

Influence of polymorphisms in VEGF, TNF-α, and GSTP1 genes on retinopathy of prematurity risk: a Meta-analysis

Summary: A meta-analysis was conducted to investigate the relationship between VEGF, TNF-alpha, and GSTP1 gene polymorphisms and ROP risk. The results showed that the VEGF -460 T/C polymorphism may be associated with a lower risk of ROP, while other polymorphisms did not show significant associations with ROP. Further research on haplotype effects of VEGF polymorphisms in ROP risk is needed.

Retinopathy of prematurity: contribution of inflammatory and genetic factors

Summary: Retinopathy of prematurity (ROP) is a significant cause of childhood visual impairment and blindness. In addition to oxidative stress, inflammation and genetic factors are also involved in ROP pathogenesis. Perinatal inflammation or infection may contribute to ROP development, and the exaggerated inflammatory response of microglia cells may promote pathological neovascularization. Treatment with angiogenic cytokines, such as anti-VEGF antibodies, has shown efficacy but some patients are refractory. Genetic factors also play a role in the phenotypic variability of ROP, but further research is needed.

Effect of Intravitreal Aflibercept vs Laser Photocoagulation on Treatment Success of Retinopathy of Prematurity The FIREFLEYE Randomized Clinical Trial

Summary: In infants with ROP, intravitreal aflibercept did not meet the criteria for noninferiority compared to laser photocoagulation in terms of treatment success at week 24. Rescue treatment was less required in the intravitreal aflibercept group, but the serious adverse event rates were higher in the laser photocoagulation group. Further data would be needed for more definitive conclusions on the comparative effects of these treatments.

Summary: This study investigated the association of single-nucleotide polymorphisms (SNPs) in VEGFA, EPAS1, BDNF, and NOS3 genes with the development and severity of retinopathy of prematurity (ROP). Significant results were found in 8 out of 11 SNPs, indicating that genetic predisposition plays a role in ROP pathogenesis and severity. Specifically, mutations in certain SNPs were associated with increased ROP severity and treatment requirement, while mutations in other SNPs were linked to milder ROP and no need for treatment. More research involving diverse populations is needed to confirm these findings and further understand the etiology of ROP.

First report of polymorphisms in MTRR, GATA4, VEGF, and ISL1 genes in Pakistani children with isolated ventricular septal defects (VSD)

Summary: This study recruited 242 subjects (121 VSD children and 121 healthy controls) from pediatric cardiac units in Lahore. The results showed associations between single nucleotide polymorphisms in MTRR, GATA4, and VEGF genes with VSD, while ISL1 gene variant did not appear to be associated with the VSD in the recruited cohort. Further validation is needed.
