Co-Inhibition of P-gp and Hsp90 by an Isatin-Derived Compound Contributes to the Increase of the Chemosensitivity of MCF7/ADR-Resistant Cells to Doxorubicin

Summary: The present study investigated the cytotoxic, chemosensitizing, and apoptotic effects of a compound called HAA(2021) on breast cancer cells and breast cancer cells resistant to doxorubicin. The study also explored the binding mode of HAA(2021) with P-glycoprotein (P-gp) and heat shock protein 90 (Hsp90) and its contribution to chemosensitization. The results showed that when combined with doxorubicin, HAA(2021) synergistically inhibited proliferation and induced significant apoptosis in breast cancer cells. It also increased the chemosensitization of resistant cells to doxorubicin. Furthermore, molecular modeling studies revealed that HAA(2021) binds to P-gp and Hsp90, leading to inhibition of the MDR efflux pump and increased chemosensitization.

Summary: This study analyzed the chemical diversity of essential oils from flowers and leaves of Achillea ligustica All. collected from different Mediterranean locations. The results showed that geographical variability influenced the compositions of the essential oils, with unique observed chemotypes and high degree of diversity among samples from different areas. Sample 8 was found to be a potential source of fragranol and fragranyl acetate, which could have applications in the insecticidal and pheromone industries.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
