Development of affinity polymeric particles for the removal of 4-dimethy-laminopyridine (DMAP) from active pharmaceutical ingredient crude streams using a green technology

Summary: Polymeric particles with affinity for 4-dimethylaminopyridine (DMAP) were developed using supercritical carbon dioxide (scCO2) technology for the cleanup of potentially genotoxic impurities from crude mixtures of Active Pharmaceutical Ingredients (APIs). These particles, known as DMAP-molecularly imprinted polymer (DMAP-MIP), showed high extraction efficiency in removing DMAP from solutions and pharmaceutical mixtures. They also exhibited high recoveries of both DMAP and the API, above 99%.

However, diafiltration still lacks a simple and compact continuous variant. A single piece of diafiltration equipment has been presented achieving continuous buffer exchange of up to 99.90%, with the vulnerability to concentration polarization significantly reduced by using an alternating direction of diafiltration buffer perfusion.

The membrane, after cross-linking treatment and synthesis of thin-film composite through interfacial polymerization, was solvent-activated to enhance permeability.
